Fragmenty pędów i zalążni izolowane z cebul niechłodzonych i chłodzonych narcyza ‘Carlton’ wykładano na pożywki inicjalne zawierające zawierające auksynę (A): Picloram lub 2,4-D w stężeniu 10-100 µM oraz cytokininę (C): BA w stężeniu 0,5-50 µM. Regulatory wzrostu zastosowano w następujących proporcjach: A > C oraz A = C. Na wszystkich eksplantatach niezależnie od rodzaju i pochodzenia obserwowano formowanie żółtego nodularnego kalusa. Proces ten zależał od proporcji auksyny i cytokininy w pożywce. Rodzaj i pochodzenie eksplantatu miał wpływ na intensywność formowania kalusa oraz na kierunek morfogenezy w kulturach uzyskanego kalusa. Najwięcej kalusa otrzymano w kulturach fragmentów podstawy pędu izolowanych z cebul niechłodzonych oraz kulturach fragmentów dolnej i wierzchołkowej części zalążni izolowanych z cebul chłodzonych. Różnicowanie zarodków somatycznych odbywa się głównie w obrębie kalusa zaindukowanego na fragmentach wierzchołkowej części pędu i zalążni, natomiast formowanie korzeni przybyszowych zachodzi w obrębie kalusa inicjowanego na fragmentach podstawy zalążni oraz na eksplantatach pochodzących ze środkowej części pędu.

Shoot and ovary pieces isolated from chilled and unchilled bulbs of narcissus ‘Carlton’ were put on initial media containing auxin (A): Picloram or 2,4-D at the concentration of 10-100 µM and cytokinin (C): BA at the concentration of 0,5-50 µM. Growth regulators were used in the following proportions: A > C and A = C. On the surface of all explants, yellow nodular callus formation was observed. The process depended on the proportion of growth regulators in medium. Explant type and origin affected the intensity of callus formation as well as direction of morphogenesis in callus culture. The highest amount of callus was obtained in the culture of lower part of shoot explants isolated from unchilled bulbs and in the culture of lower and upper part of ovary explants taken from chilled bulbs. Differentiation into somatic embryos took place mainly within the callus inducted on the upper part of shoot and ovary explants, whereas the adventitious root development in the callus initiated on the lower part of ovary explants as well as on explants originated from middle part of shoot.
