Ostatnio prowadzone badania wskazują na współdziałanie węgla i azotu w regulacji procesów metabolicznych oraz wzrostu i rozwoju roślin. Zaproponowano model, według którego geny związane z metabolizmem węgla i azotu są regulowane na zasadzie sprzężenia zwrotnego poprzez sygnały węgla i azotu. Celem badań było określenie wpływu temperatury i zróżnicowanego poziomu C : N na wzrost i rozwój pędów Clematis pitcheri rosnących in vitro. Doświadczenia przeprowadzono na wierzchołkowych fragmentach pędów Clematis pitcheri pochodzących z rozmnażania in vitro. Eksplantaty umieszczano na pożywce podstawowej Murashige i Skooga [1962] zawierającej meta-topolinę 0,2 mg·dm⁻³. Badano wpływ dwu stężeń sacharozy (10, 30 g·dm⁻³) oraz różnego poziomu KNO₃ i NH₄NO₃ (stężenie standardowe wg pożywki - 100% i zmniejszone do połowy - 50%) w procesie wzrostu i rozwoju pędu głównego oraz pędów bocznych powojnika. Kultury pędów rosły w fitotronie w warunkach 16-godzinnego oświetlenia w temperaturze 15°C, 20°C i 25°C. Obniżenie poziomu azotu w pożywce o połowę (50% KNO₃ i NH₄NO₃) oraz niskie stężenie sacharozy - 10 mg·dm⁻³ uaktywniało wzrost pędów bocznych (średnio 3,3) powojnika, ale tylko u eksplantatów rosnących w temperaturze 25°C. Najmniej pędów bocznych (0,6-0,7) powstawało w temperaturze 15°C, w obecności wyższego stężenia sacharozy (30 g·dm⁻³) i przy obydwu poziomach soli mineralnych. Wzrost stężenia sacharozy w pożywce ograniczał powstawanie i wzrost pędów bocznych, niezależnie od stosowanego poziomu soli azotowych. Najdłuższe pędy boczne uzyskano w temperaturze 20°C i 25°C, na pożywce zawierającej 100% stężenie KNO₃ i NH₄NO₃ oraz niski poziom sacharozy (10 g·dm⁻³). Pędy powojnika mnożone w temperaturze 20°C i 25°C charakteryzowały się wyższą świeżą masą niż eksplantaty rosnące w 15°C.

It is know that in plants, sugar and nitrogen are critical nutrients. Sugar can act as a signal that trigger changes in gene expression and influence the developmental and metabolic processes in plants. Nitrogen sources have been also shown to regulate gene expression. A model was proposed in which genes involved in carbon and nitrogen metabolism are cross-regulated by both carbon and nitrogen signals. The influence of different sucrose/nitrogen levels on the growth and development of Clematis pitcheri cultured in vitro was investigated. The apical stem section (2-3 nodes) were placed on MS basic culture media with m-Topolin 0.2 mg·dm⁻³. The influence of sucrose (10, 30 g·dm⁻³) and different levels of KNO₃ and NH₄NO₃ (normal strength - 100%; half strength - 50% according to the MS medium) in the process of main and axillary shoot growth and development were investigated. The cultures were incubated at the temperatures of 15°C, 20°C and 25°C. The highest number of axillary shoots (3.3) was obtained on the explants growing at the temperature of 25°C, in the presence of 50% of KNO₃ and NH4NO3 strength, and sucrose at 10 g·dm⁻³. On the other hand, the culture growing at the temperature of 15°C produced the lowest numbers of axillary shoots (0.6-0.7) on the media containing sucrose at 30 g·dm⁻³ and both strengths of KNO₃ and NH₄NO₃. High concentration of sucrose inhibited the formation and elongational of axillary shoots. The best elongation of axillary shoots was found at the temperatures of 20°C and 25°C on the medium with a normal strength (100%) of KNO₃ and NH₄NO₃ and a low (10 g·dm⁻³) concentration of sucrose. The shoots growth at the temperatures of 20°C and 25°C had a higher fresh mass than the explants cultured at 15°C.
