Wśród liliowców wiodącą pozycję zajmują obecnie tetraploidy. Ich kwiaty są niezwykle okazałe. Jednakże podczas przechodzenia z form diploidalnych do tetraploidalnych, przy użyciu kolchicyny, oprócz kompletnych tetraploidów powstają chimery - miksoploidy. Ponadto ze względu na silne działanie kancerogenne kolchicyny, coraz szerzej stosuje się antymitotyki mniej szkodliwe dla człowieka - oryzalinę, trifluralinę i amiprofos metylu. Celem badań było opracowanie efektywnej metody poliploidyzacji in vitro liliowca z wykorzystaniem alternatywnych dla kolchicyny, mniej szkodliwych dla człowieka antymitotyków. Do badań użyto ustabilizowane kultury pędów przybyszowych liliowca trzech diploidalnych genotypów: Hemerocallis multiflora, H. hybrida ‘Blink of An Eye’ i ‘Fizziwig’. Antymitotyki (mg·dm-3) - kolchicynę (500), oryzalinę (5 i 15), amiprofos metylu (5 i 10) oraz trifluralinę (50 i 100) dodawano do pożywki. Traktowano nimi kultury pędów przez 2 lub 7 dni, następnie pędy przenoszono na pożywkę stymulującą regenerację pędów (bez antymitotyków), na której utrzymywano je przez 2 lub 4 kolejne 6-tygodniowe cykle namnożeniowe, po czym je ukorzeniano i sadzono w szklarni. Po 2. i 4. cyklu namnożeniowym w zregenerowanych pędach oceniano poziom ploidalności metodą cytometrii przepływowej. Tetraploidy uzyskano odpowiednio dla każdego z genotypów: H. multiflora - 8 (13,1%), ‘Fizziwig’ - 6 (5,6%), ‘Blink of An Eye’ - 20 (15,4%). Najwięcej tetraploidów uzyskano przy zastosowaniu oryzaliny i APM. Kolchicyna w badanym stężeniu - 500 mg·dm-3 okazała się dla jednego genotypu wysoce fitotoksyczna - pędy zamarły. Efektywność uzyskania tetraploidów - średnia dla wszystkich genotypów i traktowań, wyniosła 11,4%. Wykryto także 9,4% miksoploidów.

Tetraploids have a leading position among the daylily cultivars due to the desirable traits such as vigorous growth and large flowers. However, during the induction of polyploidy using colchicine, except the solid tetraploids, the mixiploids also occur. Moreover, colchicine is toxic to humans. Therefore this antimitotic agent should be substituted by other agents, such as amiprophos methyl, oryzalin or trifluralin, which are known to be less harmful. The objective of this study was to obtain daylily tetraploids and to develop an effective method of polyploidization using an alternative to the colchicine antimitotic agents. In vitro shoot cultures of three daylily diploid genotypes were used in this study: Hemerocallis multiflora, H. hybrida cultivars ‘Blink of An Eye’ and ‘Fizziwig’. Antimitotic agents (mg·dm-3), colchicine (500), oryzalin (5 and 15), amiprophos methyl (APM) (5 and 10) and trifluralin (50 and 100) were added to a regeneration medium. Shoot clumps were exposed to the antimitotic agents for two or seven days and then transferred on the same medium without these agents. On those media, the explants were subcultured two or four times every six weeks. Then the regenerated shoots were rooted in vitro and planted in a greenhouse. Ploidy level was evaluated using flow cytometry. Tetraploids were detected for each genotype, eight (13.1%) in Hemerocallis multiflora, six (5.6%) in the cultivar ‘Fizziwig’, and twenty ones (15.4%) in ‘Blink of An Eye’. The highest numbers of tetraploids, approximately 20%, were obtained for the treatments with APM and trifluralin. Colchicine at the tested concentration revealed a highly phytotoxic effect, all explants of one of the genotypes died. The efficiency of polyploidization, an average for all genotypes and treatments, was 11.4%. There were also 9.4% mixoploids detected.
