The effect of ethanol and benzene and xylene on toluene biotransformation to benzoic acid, was investigate in rats exposed to solvents vapours by inhalation. One-, two-, and three-component solvent systems (toluene, toluene + benzene, toluene + xylene, toluene + benzene + xylene) at different concentrations (1.5, 4.5, and 13.5 mmol/m3 of air), were used. The exposure was carried out in a dynamic toxicologic chamber for four successive days, 6 hrs per day. Thirty minutes prior to every single experiment rats were treated by stomach tube with ethanol (aqueous solution) in a dose of 1366 nr 2732 mg/kg body weight (1/10 or 1/5 LD50). The controls received distilled water only. Urine concentration of benzoic acid was measured within the first and the second day after termination of the exposure, using gas chromatographic method. No effect of ethanol on urine elimination of benzoic acid was observed in animals exposed to solvents vapours at the concentration of 1.5 mmol/m3 of air as well as in the second twenty-four hours in animals put to tests under all experimental conditions applied. In animals exposed to solvents vapours at the concentration of 4.5 mmol/m3 and treated with ethanol in a dose of 1366 or 2732 mg/kg body weight, a reduction of urine elimination of benzoic acid amounting to from 15 to 28% or from 54to 41 %, respectively, was noted. On the other hand, when ethanol was administered in a dose of 1366 mg/kg body weight, in rats exposed to: toluene plus xylene or toluene plus benzene plus xylene (13.5 mmol/m3 of air), an increase of urine elimination of benzoid acid by 37% or 31/%, respectively, No found. At the higher dosing level of ethanol, urine elimination of the said toluene metabolite was increased by from 60 to 103% following the exposure to all combinations of solvents vapours under test. Results obtained allow for conclusion that reciprocal effects of ethanol (at dosing level of 1366 and 2732 mg/kg body weight) and solvents vapours (at concentration of 4.5 and 13.5 mmol/m3 of air) on urine elimination of benzoic acid, are in the nature of metabolic interactions between the xenobiotics Bed.
