Jony chromu (III), podobnie jak genisteina, mogą pełnić rolę modulującą proces proliferacji komórek. Celem przeprowadzonych badań było określenie zmian dynamiki podziałów komórkowych miocytów mysich linii C2C12 zależnych od stężeń chromu oraz genisteiny. Do badań zastosowano standardową pożywkę (DMEM) zawierającą 10% płodowej surowicy bydlęcej (FBS) poddanej uprzedniej adsorpcji związków hydrofobowych na kolumnie Lipidex-1000. Chrom dodawano do medium w postaci pikolinianu [C18H12N3O6Cr] lub octanu [(CH3C02)7Cr3(OH)2] w 5 różnych stężeniach (0.1 - 1000 µgCr3+/L). Genisteinę badano w stężeniach 1, 10 i 100 µM. Czynniki doświadczalne stosowano w okresie od 48 do 96 godz. hodowli, po czym próbki zbierano i liczono w komorze Burkera. W badaniu kontrolnym efektu oczyszczania FBS na lipideksie stwierdzono nieoczekiwany 50% wzrost liczebności komórek po usunięciu hydrofobowych związków, co wskazuje że ich obecność ma hamujący wpływ proliferacje. Wprowadzenie rosnących stężeń chromu do medium hodowlanego spowodowało proporcjonalny spadek podziałów komórek począwszy od 10 µgCr3+/L dla pikolinianu oraz od 100 µgCr3+/L dla octanu z maksymalnym efektem hamowania przy 100 µgCr3+/L (pikolinian; 80%) i 1000 µgCr3+/L (octan; 55%). Genisteina dodana do medium hodowlanego w stężeniu 1 i 10 µM powodowała tylko niewielkie zmniejszenie dynamiki podziałów (25%), natomiast stężenie 100 µM spowodowało całkowitą blokadę podziałów komórkowych (100%). Uzyskane wyniki wskazują na większą aktywność biologiczną chromu stosowanego w postaci pikolinianu aniżeli octanu. Podsumowując można stwierdzić, że obserwowane efekty działania wysokich i niskich stężeń obu badanych związków są zgodne z założeniem ich dwufazowego działania oraz, że stężenia jonów chromu do 1 µgCr3+/L oraz genisteiny do 10 µM można uznać za wartości graniczne.

Chromium (III) ions similarly to genistein may act as modulators of cell proliferation. The aim of the present study was to describe changes of mouse myocytes line C2C12 proliferation caused by different chromium or genistein concentrations. The experiments were performed using standard (DMEM) containing 10% of fetal bovine serum (FBS) delipidated on Lipidex-1000 column. Chromium was added to medium in as picolinate [Cl8H12N3O6Cr] or acetate [(CH3CO2)7Cr3(OH)2] at 5 different concentrations (0.1 - 1000 µgCr3+/L). Genistein was testes at concentrations of 1, 10 and 100 µM. Cells were incubated with experimental media from 48 to 96 hrs of cultivation. The cells were counted using Biirker chamber. In control incubations we found that delipidation of FBS resulted unexpectedly in 50% increase of cell proliferation, indicating the presence of hydrophobic inhibitory compounds in row unpurified FBS. Introduction of increasing chromium concentrations to culture resulted in proportional decrease of cell division beginning from 10 µgCr3+/L for picolinate and 100 µgCr3+/L for acetate with maximal inhibitory effect at 100 µgCr3+/L (picolinate; 80%) and 1000 µgCr3+/L (acetate; 55%). The enrichment of medium with genistein at concentrations of 1 or 10 µM caused only minor effect (>25% inhibition), while genistein concentration of 100 µM totally inhibited proliferation (100%). The results in these studies showed higher biological potency of chromium picolinate then acetate. In conclusion, the observed effects of low and high concentrations of tested compounds are in agreement with the assumption of their biphasic mode of action suggesting that critical concentrations can be considered as 1 µgCr3+/L for chromium (III) and 10 µM for genistein.
