The purpose of the study was to determine the usefulness of Gradisol G (Polfa — Kutno) in the isolation of peripheral blood lymphocytes of pregnant cows and sheep. After isolation the percentage of the individual cells was evaluated using the Thom’s chamber. The morphology, vitality and recovery of the cells compared with the total number of peripheral blood lymphocytes were also assessed. The blastic transformation test was applied to evaluate the immunologic activity of lymphocytes isolated in the optimal Gradisol G gradient. The findings were compared witr the results obtained using a Lymphoprep (Nyegaard) 1,077 gradient. The studies showed that Gradisol G fulfilled all the requirements for the succesful isolation of peripheral blood lymphocytes of pregnant cows and sheep. The optimal density to isolate peripheral blood lymphocytes of cows and sheep was 1.077 g/ml.
