Transcription is the main step in the regulation of gene expression. To study this process in vitro, it is necessary to obtain highly purified RNA polymerases. Here, we describe a method of RNA polymerase purification using a Mono Q FPLC column. Using Mono Q column chromatography accelerates the purification process and separates RNA polymerase II from RNA polymerase III with good yield.
We briefly review the limited application of marker assisted selection in past wheat breeding programmes, and contrast the current situation, where increasingly it has become feasible to tag almost any gene with a microsatellite assay. Although this capability is having an impact on the conduct of large breeding programmes, a much more profound change in breeding strategy will become possible when SNP technology has matured sufficiently so that the throughput of molecular marker-based genotyping will be able to keep pace with the numbers of plants that breeders can handle in the field. We discuss the considerations that will need to be addressed in the generation of a new breeding paradigm to take advantage of the genomics revolution.
The research draws on 1970-1998 decade rainfall, maximum, minimum and average air temperature, relative air humidity recorded at 1 p.m. and insolation; all of them reported by 6 Meteorological Office (IMGW) stations and by one agricultural meteorological station and on spring wheat yielding reports provided by Provincial Statistical Office (WUS). Multiple regression was used to define the relationship between yield and weather components over respective periods. The regression equations developed for the Zachodnie Pomorze Province can be applied to forecast spring wheat yielding. Preliminary forecasts are available late in May, whereas precise forecasts late in June and July.
A fragment of wheat nuclear DNA was shown to be able to replicate autono­mously in wheat nuclei. The fragment was 637-bp long and contained telomeric repeats at both termini. Its replication was manifested by the appearance of a radioactive reaction product of the same approximate size. Further analyses showed that the linear, double-stranded réaction product was labelled along its entire length and contained the same number of similarly located HaelR sites as did the fragment tested. Prokaryotic DNA remained twlabelled under the same assay conditions.
