The β-endoglucanase is one of the enzymes taking part in the degradation of the cell wall structural polysaccharides. The use of two Triticale varieties differing in their resistance to the preharvest sprouting allowed the comparison of that enzyme activity and changes in the internal structure of the cell wall observed in the light and electron microscope. The most interesting observations seem to be the channels and even the holes in the walls of aleurone cells found mostly in the samples showing the elevated activities of β-endoglucanase. As those samples concern the grains of a lower resistance to pre-harvest sprouting, it might be suggested that the loosening of the wall structure may be one of the probable mechanisms in facilitating the enzyme, metabolite and water translocations through the grain tissues. Those changes can accelerate the water imput into the grain, the metabolic process and thus the increased susceptibility to sprouting.
Phytoplankton community, diatom to dinoflagellate ratio and pigment composition in surface waters with nutrient data from April 2013 to March 2014 were monitored in the south-eastern (SE) Black Sea using high performance liquid chromatography (HPLC) and microscopic analyses. Microscopic examination revealed a total of 71 species that consist of dinoflagellate (58%), diatoms (25%) and other groups (17%). Microscopy and HPLC-based pigment analyses revealed almost similar results which suggest that the phytoplankton community is mainly composed of diatoms, dinoflagellates and coccolithophores. Fucoxanthin (mean 0.35 ± 019 μg L−1), peridinin (mean 0.18 ± 0.14 μg L−1) and 19-hexanoyloxyfucoxanthin (mean 0.24 ± 0.15 μg L−1) are prominent pigments which showed significant correlation with Diatom-C (r2 = 0.63–0.71, p < 0.05), Dinoflagellate-C (r2 = 0.49–0.80, p < 0.05) and Coccolithophore-C (r2 = 0.72–0.82, p < 0.05), respectively. Mean carbon biomass of diatoms (36.50 ± 9.72 μg L−1) was higher than that of dinoflagellates (33.32 ± 9.05 μg L−1). Significant differences were also observed in nutrient ratio (N:P and Si:N) (One-way ANOVA, p < 0.05). Results illustrate that HPLC-based pigment approach can be used for taxonomic characterisation of phytoplankton groups in the SE Black Sea. Moreover, relatively high dinoflagellate species dominancy and significant correlations between Phyto-C and marker pigments indicate that phytoplankton community composition is shifting towards much smaller groups in SE coasts of the Black Sea.
The objective of this study was to investigate the ascarid infection in Asiatic lions using scat samples, based on microscopic analysis, PCR amplification of the ITS-2 region of ribosomal DNA and sequence analysis of the amplicons. Microscopic analysis indicated the presence of eggs of Toxascaris leonina in eleven of the sixteen scat samples analysed and in one of these eleven scats eggs of Toxocara cati were also detected. In five of the scats eggs were not detectable. The presence of T. leonina in all the infected samples was also confirmed by PCR amplification of the ITS-2 of ribosomal RNA gene and five of these also showed amplicons corresponding to T. cati, respectively. Toxocara canis infection was not observed in any of the scat samples. Nucleotide sequence analysis of the ITS-2 region indicated 97% to 99% similarity with T. leonina and T. cati, respectively. To our knowledge, this is the first molecular characterization of ascarid infection in captive Asiatic lions from a zoological garden of India. This study also indicates that Asiatic lions are more prone to infection either with T. leonina or T. cati and the parasite is not host specific.
A single adult female specimen of Halosaurus ovenii Johnson, 1864 was captured by trammel nets at a depth of about 200 m off the coast of Arbatax (Sardinia, Italy) in early April 2007. Macroscopic and microscopic analysis of the gonad showed a postspawning ovary. This is the fourth documented capture of this fish In the Mediterranean Sea, representing the north-easternmost record for this species in this geographic area. Furthermore, the present specimen was fished at the shallowest depth ever recorded before.
