Tissue cultures were derived from the roots and above-ground part of sterile seedlings of Leuzea carthamoicles DC. Testing of the growth on the medium according to Murashige and Skoog with the addition of various combinations and concentrations of growth substances proved that at calluses derived from the aerial part the highest stimulative effect was carried out by the combination of 2,4-D (1.0 mg1-1) and IBA (0.5 mg1-1). The average value of the increment was 544%. Callus cultures derived from the roots grew less intensively than the cultures derived from the aerial parts. The highest stimulative effect was found at 2,4-D (1.0 mg1-1) in combination with BA (1.0 mg1-1) - 230%. During experiments for a certain level of morphological differentiation bud-formation was reached and after rooting plant regeneration. When substituting 1AA by Meristimul and its combination with kinetin as well, deformed buds were formed. Orientation chromatographic analysis of callus extracts and leaves of regenerated plants has not proved the presence of ecdysteroids so far.
The callus obtained from the growing plant of Ononis arvensis L. was cultivated on the Murashige-Skoog (MS) medium containing a-naphtylacetic acid (NAA) in concentration of 10 mg/1 or 2,4-dichlorphenoxyacetic acid (2,4-D) in concentration of 1 mg/l. It has been found out that it should be taken 0.5-1.5 g of the callus, for starting a long-term cultivation. In this case the duration of the lag phase is cut down. The MS medium containing NAA caused a higher increase of the biomass (the higher percentage rate of the fresh mass increase - 1500) in comparison with the MS medium with 2,4-D (the rate of the fresh mass increase - 250). Additionaly, the higher flavonoids content (0.10%) was noticed in the callus cultivated on the MS medium containing NAA at the steady light (this result refers to the 25 - 40 day callus).In the same time the tissue culture cultivated of the day-light period was still is the phase of the intensive growth. The number of passages play also an important role in the biosynthesis of flavonoids. During the first passages the flavonoids content reached the value of 0.20%, after ten passages - 0.15%, whereas after twenty passages - only 0.05%.
Na eksplantatach liścieniowych Capsicum annuum L., cv. Bryza auksyna (IAA) indukowała tworzenie tkanki kalusowej oraz korzeni. Natomiast cytokinina (BAP) stymulowała nie tylko wzrost kalusa, ale również przy najwyższym stężeniu, tworzenie pąków przybyszowych. Dla wydajnego tworzenia pąków jak i korzeni niezbędna jest obecność obu regulatorów wzrostu. Nie wykazano większego wpływu czynnika endogennego (część dystalna i proksymalna liścienia) na zdolności regeneracyjne.
Naftochinony - substancje barwne i alkaloidy pirolizydynowe były badane w tkance kalusowej i kulturze zawiesinowej Arnebia euchroma. W celu rozdzielenia frakcji oraz przeprowadzenia analizy jakościowej i ilościowej zastosowano metody chromatografii kolumnowej (CC), chromatografii cienkowarstwowej (TLC) oraz wysokociśnieniowej chromatografii cieczowej (HPLC). Najwyższą zawartość szikoniny (1850 mg 1-1) znaleziono w kulturze zawiesinowej hodowanej w pożywce Davydenkov Arnebia (DA) uzupełnionej jasmonianem metylu - MJ (100 uM). W kontroli (pożywka DA bez MJ) wykryto tylko 280 mg l-1 szikoniny. W tkance kalusowej hodowanej na pożywce DA z pikloramem (10 mg l-1) oznaczono acetyloszikoninę i ślady alkaloidów pirolizydy- nowych (PA). Pikloram zwiększał wskaźnik wzrostu kalusa lecz jednocześnie zmieniało się zabarwienie tkanki z czerwonej na żółto-zieloną. Zarówno kalus jak i kultura zawiesinowa miały 11 lat. Tkanki te przez cały okres były hodowane na pożywce Davydenkova, która w powyższej pracy została użyta do kontrolnych badań porównawczych.
The paper provides the review of research on interaction in dual cultures in vitro (fungus and plant tissue culture of its host plant) with marking test results in this area in order to show usefulness of this method in testing pathogenicity and the possibility of its use in forest practice
