The effect of 5alpha-dihydrotestosterone (DHT) on the level of human prostatic acid phosphatase (hPAP) mRNA was studied using tissue slices from various benign prostatic hyperplastic glands. The absence of DHT in the incubation medium led to a gradual, significant decrease of the hPAP mRNA level. Addition of the hormone induced hPAP mRNA in a time- and dose-dependent manner. The maximal 2-4-fold induction by 10(-9) M DHT was observed after 3-5 h of incubation, and then the hPAP mRNA level was 6-20-fold higher than that in a parallel sample incubated without DHT. The results suggest that DHT is necessary to sustain the expression of hPAP in hyperplastic prostates.
The objective of present experiment was to reveal influence of taurine, nanoparticles of gold or nanoparticles of gold conjugated with taurine on molecular and morphological status of muscle’s fibre proliferation activity at the end of embryogenesis. The expression of Proliferating Cell Nuclear Antigen (PCNA) was determined in embryonic chicken pectoral muscle using real time PCR and immunohistochemical method. Furthermore, we calculated the average number of nuclei within the muscle fibre fasciculus in cross sectional image. Taurine, supplemented in ovo to chicken embryo, enhanced organization of breast muscle by activating molecular mechanisms evaluated as an expression of PCNA. Nanoparticles of gold did not influence examined parameters.
A differential expression pattern of spermidine/spermine N1-acetyltransferase (SSAT), the enzyme critical to proper homeostasis of cellular polyamines, is reported in mouse kidney undergoing hyperplasia and hypertrophy. We have shown that SSAT activity and SSAT mRNA are significantly induced by antifolate CB 3717 and folate that evoke a drug-injury-dependent hyperplasia. In contrast, SSAT activity is down-regulated in the testosterone-induced hypertrophic kidney, while SSAT mRNA is positively controlled by this androgen. Catecholamine depletion evoked by reser- pine drastically decreases the folate-induced activity of «S-adenosylmethionine decar- boxylase (AdoMetDC), which limits polyamine biosynthesis, but has no effect on SSAT activity augmented by CB 3717. Our results document that the increased SSAT expression solely accompanies the proliferative response of mouse kidney, and sug­gest the importance of post-transcriptional regulation to the control of SSAT activity in both hyperplastic and hypertrophic experimental models.
