The immunological effects of environmental and/or occupational exposure to S02 in air as polluting gas have been examined in a group of 21 exposed men. The determination of S02 concentrations in ambient air in the work enviroment as well as in ambient air in residential areas of these men was performed, always using Amaya-Sugiura passive sampling and ion spectrophotometry. Mean concentration of S02 in ambient air in residential areas was 0.5792 mg • m-3 ± 0.2871 (0.22 to 1.52). Mean concentration of S02 in ambient air in the work enviroment was 2.2612 ± 2.1477 mg • m-3 (0.47 to 9.57). For the determination of T-cells and (CD19+)B-cell populations Behring monoclonal antibodies were used in indirect immunofluorescence tests. The serum levels of immunoglobulins: G, A, M, E; C3c and C4 complement components; total circulating immune complexes (CIC) as well as acute phase proteins: C-reactive protein (CRP), haptoglobin, ceruloplasmin and transferrin were determined by nephelometry. The mean number of total lymphocytes in men exposed to S02 was increased by 43% (p < 0.001). The stimulation T-cell line exposed to S02 was evidenced by increased number of (CD3+)T-cells, by about twice (p < 0.001) increased number of (CD4+)T-helper cells and by 68% (p < 0.001) increased number of (CD8+)T-suppressor cells. The higher increase of (CD4+)T-helper cells compared to (CD8+)T-suppressor cells caused the increased value of the (CD4+)T-helper/(CD8+)T-suppressor ratio by about 25% (p < 0.01) in the subjects exposed to S02. In contrast, a decreased number of (CD16+)NK cells by about 38% (p < 0.001) in men exposed to S02 was also observed. No changes were observed in the number of (CD19+)B-cells, as well as in the (CD3+)T/(CD8+)T-suppressor ratio. In the group exposed to S02, elevation of IgG serum levels by 23.3% (p < 0.001) was evidenced, as well as a decrease of C3c by 15.6% (p < 0.001) and C4 by 30% (p < 0.001), whereas total CIC in the serum was elevateds in same conditions by about 74% (p < 0.001). No changes were observed in the serum levels of IgA, IgM, IgE, CRP, haptoglobin, ceruloplasmin and transferrin. Significant positive correlations Irl (0.51 to 0.57) were only between S02 concentrations in air and serum concentrations of total CIC. Moreover, significant negative correlations Irl between S02 concentrations in air and leucocytes (WBC), and the determined T-cells populations and (CD19+)B-cells also were observed. In conclusion, environmental exposure to S02 can enhance proinflammatory processes in degree of exposure dependent manner and also can change some parameters of cell-mediated immunity.
The experimental material comprised 20 adult Wistar rats aged 12 weeks, divided into two equal groups (control and experimental) including five males and five females each. From the first day of the experiment, the experimental group was fed Murigran feed supplemented with ß-l,3/l,6-D-glucan (Biolex Beta-HP) at a dosage of 12-19 mg/rat/d, depending on body weight, while the control animals were administered the same feed without any additives. On days 1 and 14 of the experiment, arterial blood samples were collected and diluted with heparin, and then the following parameters were determined: total protein and γ-globulin contents, lysozyme and ceruloplasmin activities, the proliferative response of blood lymphocytes after stimulation with LPS or ConA, metabolic activity, and the potential killing activity of phagocytes. The results showed that Biolex-Beta HP modulated the selected parameters of specific and non-specific humoral and cellular immunity in rats.
The aim of the study was to determine the degree of phagocytic activity of neutrophils, lymphocyte histogenesis and total immunoglobulin level in six bitches with glandular cystic hyperplasia - pyometra complex (GCH - PC) before and after ovariohysterectomy in comparison to healthy dogs. Haematological study showed significant increases (P<0.005) in the white blood cell count and neutrophilia. After ovariohysterectomy these parameters gradually returned to normal. The phagocytic ability (expressed as phagocytic activity PA and index of phagocytic activity IPA) of neutrophils was impaired in bitches wit GCH - PC and has improved during the therapy. The degree of concanavalin A induced lymphocyte blastogenesis expressed as a stimulation index was significantly (P<0.005) depressed in affected bitches and after surgical treatment had not reached values comparable with that found in healthy dogs. The level of total immunoglobulins was significantly (P<0.05) decreased at the beginning of observation and gradually increased after treatment. The results of the present study have provided usuful information for diagnosis and prognosis of GCH - PC in bitches because the success of the treatment and recovery from the GCH - PC is in direct relation with the optimal condition of the immune system.
The experiment was performed on 32 suckling Kamieniec lambs divided into two equal groups: I - control and II - experimental. Over a period of 60 d, experimental lambs were fed a diet supplemented with Saccharomyces cerevisiae yeast with an increased ß-l,3/l,6-D-glucan and mannan-oligosaccharide content. The following meat performance parameters were determined: body weight, daily gains, growth rate, the dimensions of musculus longissimus dorsi sections, skin and subcutaneous fat thickness, and fat thickness over the loin „eye“ area. Serum humoral immunity parameters: lysozyme and ceruloplasmin activities, total protein, and γ-globulin contents were determined. The yeast supplement significantly increased the values of the analysed meat performance traits. As regards the investigated humoral immunity parameters, i.e. lysozyme activity, ceruloplasmin activity (throughout the entire experiment), and γ-globulin content (from experimental day 30 to 60), significantly higher values were noted in the experimental group, compared with the control group. Statistically significant differences between the compared groups were not observed only with respect to total protein content.
The objective of this study was to determine the stimulating effect of the brewers' yeast Saccharomyces cerevisiae dietary supplement on selected parameters of specific and non-specific humoral and cellular immunity in lambs. The study involved 32 lambs aged 30 ±3 d, divided into two equal groups: control and experimental. Animals in the experimental group were fed a C-J concentrate mixed with a prebiotic, the extract of dried brewers' yeast containing 10%-15% MOS and 25%-30% ß-l,3/l,6-D-glucan in the amount of 3 g/kg of the concentrate. At the beginning of the experiment (day 0) and on the 15th, 30th, and 60th d of the study, blood was sampled from the jugular vein to determine γ-globulin levels, lysozyme and ceruloplasmin activity, proliferative response of blood lymphocytes (MTT) after stimulation with LPS or ConA, metabolic activity (RBA), and potential killing activity (PKA) of phagocytes. As regards humoral immunity parameters, significantly higher γ-globulin levels and higher lysozyme and ceruloplasmin activity were noted in the blood serum of experimental lambs supplemented with the yeast extract, in comparison with control lambs not fed the supplement. No statistically significant differences in serum total protein were found between the control and experimental groups. The analysis of cellular immunity indicators revealed significantly higher levels of RBA and PKA, and higher MTT rates after stimulation with LPS or ConA in the experimental group, in comparison with the control group.
The aim of the study was to assess bacteriolytic activity of lysozyme and serum amyloid A (SAA) level in sheep affected with zearalenone mycotoxicosis. Bacteriolytic activity of lysozyme, and serum amyloid A (SAA) level are both the elements of innate humoral immunity. Lysozyme bacteriolytic activity in serum was determined by diffusion-plate method with reference to Micrococcus luteus. SAA concentration was determined by the use of commercial ELISA kit (Phase Serum Amyloid A Assai TP802). The highest lysozyme bacteriolytic activity was observed in sheep with total rectal and intestinal mucosal membrane prolapse. Lysozyme concentration values in animals with partial rectal prolapse were significantly lower. In the control group, lysozyme concentration was low and within the range considered as physiologically normal. The highest levels of SAA, which ranged from 38.5 to 172 µg/mL, were detected in sheep in which the highest lysozyme bacteriolytic activity was noted. From the data obtained, it is undoubtful that changes in SAA level in affected sheep and control sheep are reliable indicators of zearalenone mycotoxicosis progress and termination. The highest levels of SAA were noted in animals, which had the total rectal and intestinal prolapse in course of zearalenone mycotoxicosis, and after returning the rectum to the normal position, mucosal membrane necrosis occurred, which resulted in perforation and peritoneum inflammation. The observed differences in SAA level forming range in particular animals from infected group are conditioned by the character of inflammation process and intensity in organs altered by the disease, which are principal factors inducing changes in its concentration. The level of this protein in serum reflects activation state of immunological system and could be one of the criterions in sheep health assessment.
This study was conducted to identify the differences and similarities among three Japanese quail (JQ) lines (JQ lines: white, brown and wild-black) and the European quail (EQ). The qRT-PCR was used to determine the expression of growth related genes: growth hormone (GH) and Insulin-like growth factor I (IGF-I), immune genes (Interleukins 1β, IL-1β, Interferon-α, IFN-α), pigmentation genes; dopachrome tautomerase, (Dct) and endothelin receptor type B2, (EdnrB2) in several quail tissues, while PCR-RFLP analysis of the mitochondrial 12S rRNA gene was conducted in quail meat. Expression levels of the pigmentation related genes (Dct and EdnrB2) were significantly higher (P<0.05) in the JQBr and EQ lines than in JQwh and they were comparable between JQbr and wild EQ. Expression levels of the growth related genes (GH and IGF-1) were significantly higher in 3 JQ lines than in EQ. No differences between all 4 quail lines were found in the expression of the immune related genes. In conclusion, the PCR-RFLP method may be used to distinguish between the Japanese and the European quail, which is important for breeding programs, labeling meat products and biodiversity studies.
Monitoring the condition of fish and protecting them from diseases are two of the most important aspects of prevention in intensive fish culture. Nonspecific cellular and humoral defense mechanisms were studied for the first time in pikeperch (Sander lucioperca) grown in an intensive system of culture using two sizes of fish weighing approximately 50 g and 500 g. The respiratory burst activity and potential killing activity of blood phagocytes and pronephric macrophages were measured in each group. The proliferative response of lymphocytes stimulated by mitogen ConA and LPS was also examined in the two groups of fish. The lysozyme and ceruloplasmine activities in the plasma and the total protein with Ig levels in the serum were also determined. These results will be used in controlling the influence of diets and xenobiotics on nonspecific cellular and humoral defense mechanisms and for infectious disease prevention in pikeperch raised in different systems of culture.
