Decontamination effectiveness of ammonium-ferric(III)-hexacyanoferrate(II) (AFCF) in reducing the radiocaesium transfer to hen eggs was examined. The albumen activity concentration for ¹³⁷Cs was higher than that in egg yolk. The most effective reduction of radioactivity concentrations in all egg components was noted in birds treated with hexacyanoferrate from the first day of radioisotope administration. Radioactivity concentrations in hens treated simultaneously with ¹³⁷CsCI and a caesium binder were lower by >90% than those in the controls at selected measurement intervals. The decontamination efficiency lowered with the delayed AFCF treatment.
NaCl-induced aggregation of hen egg white lysozyme (HEWL) was monitored by NMR spectroscopy. Small, but significant, changes induced by salt addition in TOCSY spectra were attributed to the effect of local reorganization of protein backbone upon ion binding. Salt-induced variations in HN and Hα chemical shifts were mapped on the HEWL 3D structure which allowed the construction of a scheme of the spatial localization of potential ion binding sites. It was found that in a 0.5 M NaCl solution six chloride anions and at least one sodium cation are bound to preferred sites on the HEWL surface.
AMP-deaminase from hen stomach smooth muscle was isolated and physico-chemi­cal properties of the purified enzyme were investigated. The enzyme had an activity optimum at pH 6.5, and poorly deaminated the substrate analogues tested. At optimum pH (6.5), in the absence of regulatory ligands (control conditions), the enzyme manifested hyperbolic substrate-saturation kinetics with half-saturation constant GS0.5) of about 4.5 mM. Additions of adenine nucleotide effectors (ATP, ADP) activated the enzyme strongly at all the concentrations tested, diminishing sig­nificantly the value of S0.5 constant. In contrast, the regulatory effect of ortho­phosphate was variable, and depended on the orthophosphate concentration used. The molecular mass of the enzyme subunit determined in SDS/PAG electrophore­sis was about of 37 kDa. The obtained results suggest that in different types of hen muscle, similarly as in humans and rats, expression of AMP-deaminase is under the control of independent genes.
The objective of the experiment was evaluation of some physical traits of egg quality in hens originating from Polish egg-type pedigree farms and destined for small-scale farming as well as in a breed of preserved chickens. 30 eggs from each of the three genetic groups (parental P55 Barred Rock strain, VH43 parental cross of V44 Rhode Island Red and H33 Leghorn strains, and the preserved Z11 Partridge Greenleg strain) were taken to analysis. The layers were maintained in the same environmental conditions and were under the same feeding regime.Analyses of egg quality were accomplished in 26th and 50th weeks of age, with an electronic EQM device. Egg weight in VH43 cross in the beginning of laying was 61.5 g and it was significantly bigger than in P55 hens (50.2 g) and Z11 hens (42 g). No statistical differences between experimental groups were found in shell thickness, shell density. Egg shell colour in P55 hens differed significantly from that in the other groups. The highest Haugh unit score (99.9) and albumen height (9.7 mm) were noted in the eggs of P55 hens in 26th weeks of age. These parameters lowered with age in all experimental groups. The highest proportion of yolk was found in the eggs of Z11 (29.5%) in 50 weeks of age. Yolk percentage increased with age in all genetic groups. Yolk colour index (La Roche) diminished with age of layers.
Four immunostimulating preparations (IPs) – Levamisol, Lidium KLP, Echinacea and Baymix Se+E – were administered to Hy-Line hens aged 8 months (group I, II, III and IV, respectively, 10 birds in each). The egg white lysozyme activity (LA) was determined before administering the IP, and next 17, 24, 31, 45 and 60 days after. All IPs led to an increase in the egg white LA which was maintained over a period of 45 (group II, III and IV) and even 60 days (group I) after administration. Levamisol was shown to be most effective IP, while Baymix Se+E the least.
A new approach has been developed to study binding of a ligand to a macromolecule based on the diffusion process. In terms of the Fick's first law, the concentration of free ligand in the presence of a protein can be determined by the measurement of those ligands which are diffused out. This method is applied to the study of binding of methyl-orange to lysozyme in phosphate buffer of pH 6.2, at 30°C. The binding iso­therm was determined initially, followed by application of the Hill equation to the data obtained, then binding constant and binding capacity were estimated.
