Ageing of rural population in Mazovian province

The paper presents statistical and spatial analysis of ageing of rural population in Mazovian Voivodship. The analysis using population statistics from the Central Statistical Office of Poland, carried out on a local LAU 2 level, where rural areas were defined according to DEGURBA classification, and using classical measures of ageing proves that all rural LAU 2s in the region are old by UNO criteria, however those adjacent to cities and Ostrołęka town are relatively younger. Population of Warsaw is older than population of adjacent rural communes. In case of rural LAU2s there is a strong correlation between the share of the old and Sauvy’s index, while there is no linkage between the share of old and double ageing index.
The study of human disorders known as premature aging syndromes may provide insight into the mechanisms of cellular senescence. The main feature of cellular senescence in vitro is cessation of cell proliferation. Down syndrome (DS) and neuronal ceroid-lypofuscinosis (NCL) are clinically characterized by the premature onset of numerous features normally associated with human aging. Phytohemagglu- tinin stimulated lymphocytes derived from DS subjects showed a statistically significant diminished proliferation capacity in comparison with lymphocytes derived from NCL and healthy individuals. We demonstrated, by applying the electrophoretic mobility shift assay, slightly impaired AP-1 DNA binding activity in NCL lymphocytes and strong in DS ones. Our results showed that the same molecular mechanisms of proliferation cessation could exist in fibroblasts characterized by replicative senescence and in lymphocytes derived from individuals with premature aging syndromes (Down).
In order to improve the physiological quality of aged broccoli seed (Brassica oleracea var. Waltham29), in this study a variable magnetic field (VMF) of 3.6 mT, 60 Hz by 5min was used as pre-sowing treatment. The accelerated aging treatment was applied for 0, 48 and 72 h (AA0h, AA48h and AA72h) and then the VMF treatment, under a randomised complete block design with 4 replications of 30 seeds. For seeds aged for 72 h a final germination of 20% was obtained, whereas without magnetic stimulation it was null. However, the magnetic bio-stimulation in 48 h aged seeds was significantly negative, reducing the final germination by 45.83% compared to the treatment without magnetic field, while the VMF effects in unaged seed were zero. The variable magnetic field effects depended on the seed aging time, resulting in positive, negative and null outcomes (responses) in vigour variables.
There are many theories of aging and a number of them encompass the role of mitochondria in this process. Mitochondrial DNA mutations and deletions have been shown to accumulate in many tissues in mammals during aging. However, there is little evidence that these mutations could affect the functioning of aging tissues.

Regulation of gastrointestinal mucosal growth during aging

The increase in the aging population has led to a growing interest in achieving a better understanding of the aging process and of diseases that are predominantly expressed during advancing age. Since the structural and, in turn, the functional integrity of the mucosa of the gastrointestinal tract (GI) are maintained by constant renewal of cells, a detailed knowledge of the events that initiate and regulate mucosal proliferative processes is essential for a better understanding of the normal aging process as well as age-associated dysfunctions, including malignancy that represent disorders of tissue growth. In Fischer-344 rats, aging is associated with increased mucosal proliferative activity in much of the GI tract. On the other hand, the functional properties are either decreased or remain unchanged during advancing age. Basal gastric acid and pepsin output decline during aging, as is gastrin secretion. In contrast, antral gastrin levels increase during this period, as is mucosal histidine decarboxylase activity. The age-related decline in gastrin secretion could partly be attributed to a higher ratio of somatostatin (D) to gastrin (G) cells in the antral mucosa. The age-related rise in GI mucosal proliferative activity could not be attributed to the trophic action of either gastrin or bombesin, since they caused no significant change in mucosal proliferation in aged rats. On the other hand, EGF and TGF-alphalpha appear to be involved in regulating mucosal proliferation during aging. Aging is associated with increased activation of EGF-receptor (EGFR), the common receptor for EGF and TGF-alpha. This could be due to (a) increased levels of membrane-bound precursor form(s) of TGF-alpha resulting in increased activation EGFR signaling processes through an autocrine/paracrine mechanism, (b) heightened sensitivity of mucosal EGFR to EGF and TGF-alpha such that comparatively lower levels of these peptides are required to activate EGFR in aged than in young animals and/or (c) loss of EGFR regulatory factor(s) such as ERRP (EGFR Related Protein), a "negative regulator" of EGFR.
The article presents selected elements of studies on factors influencing the quality of life of elderly people vs the EU-27. The focus is on the multitude and diversification of the income of old age pensioners’ and disability pensioners’ households in reference to groups with other sources of income. The article also discusses the problems of the threat of poverty and the subjective sense of poverty. It also indicates the paradoxes of the financial situation of old age pensioners’ and disability pensioners’ households in Poland, which become the source of income or improvement of the situation of entire families, especially in agriculture and in rural areas, although they have one of the lowest income levels in Europe.
In this study we examined the potential role of atherosclerosis in the main arteries supplying blood to the carotid body in the organ’s morphological degradation with age. We addressed this issue by comparing the ultrastructural picture of carotid bodies and of fragments of the carotid artery bifurcation in two age-extreme groups of rats: young - 3 months old and senescent - 24 months old. Tissues were excised under surgical anesthesia, fixed in aldehydes, and processed for transmission electron microscopy. We found that the old carotid body parenchyma exhibited profound degenerative changes. Chemoreceptor cells were at various stages of atrophy, ranging from swollen mitochondria and fewer secretory vesicles to dark dehydrated cells. In contrast, the senescent carotid artery bifurcation was little different from that in young rats. Particularly, endothelial cells were in perfect condition. There were some changes in deeper arterial wall layers such as breaks in the continuity of elastic bands or a subtly different phenotype of smooth muscle cells. No foam cells or calcium build-ups were found in the arterial walls. Such changes correspond to the process of arterial wall stiffening in old age rather than to the outright atherosclerosis. Lack of atherosclerosis in the common carotid arteries, which could hamper blood flow, argues against its playing a role in the morphological age-changes in the carotid bodies.
Astrocytes are glial cells prone to morphological changes associated with age. The aim of the study was to investigate the immunoreactivity of glial fibrillary acidic protein (GFAP) in astrocytes of the periaqueductal gray matter (PAG) of the midbrain in adult and old male rats to demonstrate morphological changes associated with age and to assess morphometrically the number of astrocytes and the digital immunostaining intensity of the examined protein in PAG astrocytes of both groups of animals. In the study, 10 male Wistar rats in two age groups were used. The first group consisted of five 100-day-old animals, whereas the second comprised five 3-year-old rats. After euthanasia, the midbrain, containing PAG, was collected and embedded in paraffin blocks. Immnunohistochemical peroxidase-antiperoxidase reaction was carried out on coronal tissue sections with the use of the specific primary antibody against GFAP, goat anti-mouse IgG, peroxidase-antiperoxidase complex, and diaminobenzidine chromogen. GFAP-immunopositive PAG astrocytes were observed under a light microscope and subjected to morphometric analysis to determine their number and digital immunostaining intensity for the protein examined. GFAP-immunoreactive PAG astrocytes in 100-day-old rats showed uniform distribution. Numerous processes branching into secondary ones protruded from intensely GFAP-immunostained stellate cells. In contrast, in 3-year-old rats a significantly lower number of glial cells of different morphology was observed compared to young animals. Astrocytes had fewer primary processes without secondary branches. Morphometric analysis confirmed microscopic observations. Our findings indicate that PAG astrocytes are prone to quantitative and morphological changes with age, which, in turn, can cause disorders in emotional, pain, and defensive reactions.
The age-related inverse relationship between gene expression of lipogenic enzymes and leptin gene expression as well as inhibitory effect of leptin on lipogenic enzyme’s gene expression suggests that leptin could be responsible in part for the low rate of lipogenesis in white adipose (WAT) of old rats. Based on the data published recently we propose a model for the direct inhibitory effect of leptin on lipogenesis. This model may explain the age-related decrease of lipogenic activity in WAT. It is likely that despite of higher concentration of noradrenaline (which inhibits leptin gene expression in WAT) in old animals, the age-dependent decrease of b-adrenergic receptor density in rat adipocytes may lead to the increase of leptin gene expression and to the increase of WAT leptin concentration. High concentration of leptin in adipose tissue decreases sterol regulatory element binding protein–1 (SREBP-1) gene expression by paracrine and/or autocrine action on adipocyte, which leads to the decrease of SREBP-1c level (mature form). The suppression of SREBP-1c synthesis causes a decrease of lipogenic enzyme’s gene expression which consequently results in lower rate of fatty acid synthesis in WAT. This model does not exclude the indirect, via hypothalamus (by decreasing food consumption), inhibitory action of leptin on WAT lipogenesis. Therefore, it is likely that leptin exerts its inhibitory effect on WAT lipogenesis both directly at the level of adipocytes, and indirectly through hypothalamus by decreasing food intake. The inhibitory effect of high leptin concentration on lipogenesis in WAT of old rats could prevent over-accumulation of triacylglycerols in adipocyte, and by this way could protect against further development of the fat mass.
