Trichoderma is one of the most exploited biocontrol agents for the management of plant diseases. In biocontrol ecology, the critical factors are detection, and the monitoring and recovery of specific biocontrol agents either naturally present or deliberately released into the environment. Protoplast fusion is an appropriate tool for the improvement of biocontrol Trichoderma strains. Protoplast isolation from Trichoderma harzianum was achieved using 24 h culture age, 6.6 mg/ml Novazym L 1412 at 30°C which resulted the maximum protoplast yield of 5 × 108/ml. The self-fused protoplasts were regenerated and 12 fusants were selected based on their growth rate on 2% colloidal chitin medium. Next, a comparison was done for chitinase and antagonistic activity. Transcriptomic analysis based on quantitative real-time RT-PCR, demonstrated that T8-05 fusant expressed 1.5 fold of chit42 transcript as compared with the parental line. This fusant with 7.02±0.15U chitinase activity showed a higher growth inhibition rate (100%) than the parent strain, against Rhizoctonia solani. To obtain a genetically marked fusant that can be used as a biomonitor, the fusant was cotransformed with the gfp and amdS genes. The morphology and viability of selected cotransformant (FT8-7MK-05-2) was similar to the parent. Green fluorescing conidia were observed within the first 2 days of incubation in the soil, and this was followed by the formation of chlamydopores after 60 days. The colonisation of the gfp-tagged fusant was also monitored visually on R. solani sclerotia by scanning electron microscopy. Production of gfp-tagged fusant of Trichoderma spp. provides a potentially useful tool for monitoring hyphal growth patterns and the population of biocontrol agent isolates introduced into environmental systems.
W przeprowadzonym doświadczeniu badano wpływ temperatury inkubacji na wzrost grzybni, kiełkowanie zarodników oraz indywidualny efekt biotyczny (IEB) grzybów Trichoderma harzianum, T. pseudokoningii i T. viride. Temperatury w zakresie 5-15°C hamowały wzrost grzybni i tworzenie strzępek kiełkowych badanych izolatów Trichoderma spp. Również wartość IEB zależała od temperatury. Wszystkie izolaty Trichoderma spp. silnie ograniczały wzrost patogenów tylko w temperaturze 20°C.
Effect of non-volatile metabolites of Trichoderma harzianum together with certain salts containing Mg++, Fe+++, Mn++, Cu++, Al+++, Ca++, K++, Na+, PO4--- and SO3--- on the production and length of rhizomorphs of Armillaria borealis, A. gallica and A. ostoyae was studied. In pure medium, T. harzianum exhibited stimulating effect on rhizomorphs of A. borealis (both number and length) and A. ostoyae (only initiation). Cu++ salt totaly inhibited the initiation of rhizomorphs of Armillaria borealis, A. gallica and A. ostoyae. Effect of other compounds on the activity of T. harzianum depended on Armillaria species. The majority of chemical compounds tested supressed the activity of non-volatile metabolites of T. harzianum. Evident stimulating effect was observed under influence of sulphate salts consisting Al++ and Fe+++ on the rhizomorph number of A. borealis and A. gallica, respectively.
W pracy przedstawiono wyniki badań wzajemnych interakcji pomiędzy trze­ma szczepami grzybów strzępkowych z rodzaju Trichoderma a dziewięcioma szczepami patogennych grzybów strzępkowych. Testy biotyczne dwuorganizmowe przeprowadzono w podłożach imitujących środowisko naturalne (zmielone ziarna czterech najpopularniej­szych zbóż), co jest modyfikacją standardowo wykonywanych testów. Poszczególne mikro­organizmy testowane i testowe wykorzystywały w różnym stopniu zboża jako źródła skład­ników odżywczych, co przejawiało się niejednakowym tempem wzrostu oraz różnicami we wzajemnych oddziaływaniach. Najsilniejsze zdolności antagonistyczne wykazywał szczep Trichoderma harzianum T33, w wielu przypadkach powodując całkowitą inhibicję wzrostu szczepów testowych. Żaden z badanych fitopatogenów nie wpłynął negatywnie na wzrost szczepu T33, co zaobserwowano w interakcjach z pozostałymi szczepami testowanymi.
In vitro test of antagonistic activity of culture filtrates from Trichoderma harzianum Rifai and Trichoderma pseudo-koningii Rifai strains against post-harvest pathogens of some fruits were investigated. The undiluted culture filtrates of the two Trichoderma species completely inhibited germination of conidia/spores of all the rot pathogens, but 50% dilution showed varying degree of inhibition of spore germination. T. pseudo-koningii culture filtrate had a rather moderate to strong inhibitory effect on mycelia of the pathogenic fungi. The highest per cent inhibition of 45.6% of mycelial growth was recorded for Aspergillus niger Tiegh.
The strains of Trichoderma harzianum were assessed for their effect on chickpea growth and control of charcoal rot caused by Macrophomina phaseolina in greenhouse assay. T. harzianum strain 25-92 significantly increased the fresh and dry weights by 50-63% and 24-42%, respectively, whereas strain 29-92 increased the fresh weight of chickpea cv. Radhey and Vishwas by 12-30% but not the dry weight in the absence of M. phaseolina. A marked increase in root length was caused by both the strains. In M. phaseolina infested pots, number of lateral roots and branching decreased with nonsignificant change in weight. Significant (P = 0.05) reduction in charcoal rot disease was observed in the pots amended with T. harzianum at all the concentrations. Moreover, 60-40% reduction in disease was recorded after 14 and 28 days in chickpea varieties Radhey and Vishwas. The resistant variety Vijay does not show significant disease. The reduction in disease was more pronounced at higher inoculum concentrations of T. harzianum (107-108 cfu/g). Overall, Trichoderma strain 25-92 improved plant growth and reduced damage in presence of the pathogen. Besides disease control the growth promoting properties of the strain improve the efficacy for commercial application.
Verticillium wilt caused by Verticillium dahliae, is one of the most devastating diseases in pistachio orchards in the world including Iran. In search for an eff ective non-chemical strategy for the management of this disease, we evaluated the biocontrol potential of Trichoderma harzianum isolates obtained from the rhizosphere of healthy pistachio trees in diff erent locations of the Kerman province of Iran against V. dahliae under laboratory and greenhouse conditions. Dual culture tests in the laboratory were conducted in a completely randomized design using 72 T. harzianum isolates. Twenty isolates showed the highest in vitro antagonistic activity. Th e results indicated that all 20 isolates were capable of inhibiting the mycelial growth of V. dahliae signifi cantly. Among them, isolates Tr8 and Tr19 were the most eff ective by 88.89% and 85.12% inhibition, respectively. Extracted cell free metabolites of all eff ective isolates also inhibited the growth of V. dahliae in the culture medium signifi cantly. According to the results, isolates Tr4 and Tr6 inhibited fungal pathogen growth by 94.94% and 88.15% respectively, through production of non-volatile metabolites. In the evaluation of volatile metabolites, isolates Tr5 and Tr4 were the most eff ective by 26.27% and 24.49% growth inhibition, respectively. Based on the results of the in vitro experiments, the fi ve most eff ective isolates were selected for evaluation under greenhouse conditions for their biocontrol potential in controlling Verticillium wilt of pistachio. Results of the greenhouse, (in vivo) experiments were positive and indicated that the occurrence of wilt disease in plants treated with the antagonists alone or in combination with pathogenic fungus was lower than in plants inoculated with pathogen alone. Th e overall results of this study suggest that Trichoderma fungal antagonist may be an eff ective biocontrol agent for the control of Verticillium wilt of pistachio.
