Background. In the present study the antioxidant properties were investigated of the medicinal plants Salvia officinalis L. (Labiaceae family) and Viscum album L. (Loranthaceae), both of them known for a long time as a remedy in the traditional medicine. The aim of this study was to prove the efficiency of ultrafiltration process for the concentration of herbs extracts and to evaluate the concentrate‟s antioxidant activity. Material and methods. The extracts were prepared by maceration, using different sol-vents. After filtering the extract through Isolab quantitative filter paper “medium”, each of the filtrates was processed by microfiltration (MF; Millipore filters with 45 μm), followed by ultrafiltration (UF). The regenerated cellulose (Millipore), polysulfone and polyacrylonitrile ultrafiltration membranes were used in the experiment. The initial extracts and samples of permeate and retentate after ultrafiltration of extracts have been characterized by determination of the protein total and total phenolic content. Standard methods like ABTS and DPPH assay are used to measure the antioxidant activity. Results. For the three types of tested membranes: Millipore, PSF and PAN, PAN membrane proves to have the greatest efficiency since it shows the highest permeate flux and the greatest retention degree for bioactive compounds. The concentrated extracts obtained after ultrafiltration with polyacrylonitrile membrane had the strongest scavenging activity for all extracts. Conclusions. The results of this study has revealed that the concentrated extracts have a very high radical scavenging activity (TEAC values for sage hydro-alcoholic concentrated extracts in range 351.87-479.04 μmol Trolox/mL extract and for mistletoe concentrated extract E2 in range 345.14-426.18 μmol Trolox/mL extract; the DPPH inhibition values was over 85% for S. officinalis concentrated extracts and ranges between 66.2% and 88.2% DPPH inhibition for V. album concentrated), therefore can be considered as a good source for further medicinal applications.
The study with sage (Salvia officinalis L.) cv. ‘Bona’ was conducted in the years 1997–1999. The effect of time of harvest on yield of fresh herb sage and yield of air dry herb, yield of air dry leaves sage. The experiment was conducted with sage two-year old. Sage transplants were planted in distance 30×40 cm. The plant material was harvested twice – in May and in September. Harvest date hat significant influence on the weight of fresh sage herb. Yield of fresh sage herb in September was bigger (190.2 dt ha⁻¹) than yield of fresh sage herb in May (104.7 dt ha⁻¹). The percentage quantity of the essential oil in the herb and leaves of sage was depended on the harvest date. The herb and leaves of sage in September had bigger the percentage quantity of the essential oil than in May. The percentage quantity of the essential oil in the leaves of sage, witch was collected in September amounted above 1.5%.
Oceniono 4 uprawne populacje szałwi lekarskiej (Salvia officinalis L.) oraz formę botaniczną alba pod względem cech morfologicznych, plonowania oraz zawartości olejku eterycznego i jego składu chemicznego. Badane populacje różniły się wysokością i pokrojem roślin oraz wielkością, kształtem i barwą liści. Najwyższy plon ziela i olejku eterycznego uzyskano z populacji amerykańskiej i holenderskiej. Najwyższą zawartość olejku eterycznego w zielu (ok. 1,8%) stwierdzono u populacji czeskiej i formy botanicznej alba. Olejek tej ostatniej formy charakteryzował się największą liczbą składników (46) i najwyższym procentowym udziałem tujonu (40%).
This report describes the effect of triacontanol on shoot multiplication and production of antioxidant compounds (carnosic acid, carnosol and rosmarinic acid) in S. officinalis cultures grown on MS basal medium (agar solidified medium supplemented with 0.1 mg l-1 IAA, 0.45 mg l-1 BAP). It was found that shoot proliferation significantly increased when triacontanol at concentrations of 5, 10 or 20 µg l-1 was added to the medium. HPLC analysis of acetone and methanolic extracts of sage shoots showed that the production of diterpenoids, carnosic acid/carnosol ratio, as well as, contents of rosmarinic acid were also affected by the treatment with triacontanol. The highest stimulation effect of triacontanol was observed on the production of carnosol, where the treatment with 20 µg l l-1 increased the content of this diterpenoid 4.5-fold compared to that in the control (sage shoots growing on MS basal medium, only).
The shoots of Salvia officinalis growing in MS liquid medium supplemented with IAA 0.1 mg l-1) and BAP (0.45 mg l-1) were treated with methyl jasmonate (MeJA) to increase production of compounds with antioxidant activity (carnosic acid, carnosol and rosmarinic acid). The increase in metabolite production depended on MeJA concentration, the period of exposure to elicitor and type of compound. The MeJA action was observed 24 h after elicitation. It was found that the maximum level of diterpenoids, calculated as the sum of CA and Car (about 8 mg g-1 dry wt) was achieved at 3 days after elicitation with 20 µM methyl jasmonate. The highest amount of rosmarinic acid (about 41 mg g-1 dry wt) was achieved with 50 or 100 μM methyl jasmonate on the 5th day after elicitation. It was almost 2-fold higher compared to the control (cultures treated with only ethanol).
Przeprowadzono ścisłe, trzyletnie doświadczenie nad wpływem sposobów zakładania plantacji, terminu zbiorów surowca i wysokości cięcia roślin na plon i zawartość olejku w Hb Salviae officinalis L. z uprawy jednorocznej. Doświadczenie zakładano corocznie przez wysiew nasion wprost do gruntu, przez wysadzanie sadzonek z inspektu oraz wysadzanie sadzonek z rozsadnika. Rośliny ścinano na surowiec w sierpniu, wrześniu i październiku między 1-2 węzłem oraz między 3-4 węzłem. Stwierdzono, że najwyższy plon surowca otrzymuje się zakładając plantację szałwii metodą bezpośredniego siewu nasion do gruntu, zbiorze w połowie października i na wysokości cięcia pomiędzy pierwszym a drugim węzłem. Natomiast największą procentową zawartość olejku stwierdzono w Klęce w sierpniu (1,30%) i we wrześniu w Szczekarkowie (1,66%). Na zawartość olejku w zielu szałwii mają wpływ: termin zbioru surowca oraz warunki klimatyczne i edaficzne, anie sposoby zakładania plantacji.
Oddzielnie dla każdego gatunku ziół założono doświadczenie poletkowe metodą bloków losowych. Czynnikiem doświadczalnym były trzy rodzaje oprysku dolistnego roślin kontrola (woda destylowana) oraz jednorazowy i dwukrotny oprysk preparatem „Multivit plus" o stężeniu 0,1%. W czterech fazach rozwojowych oznaczano akumulację suchej masy w fazie szybkiego wzrostu I, w fazie szybkiego wzrostu II, w początku kwitnienia (dla szałwii początku wzrostu liści), w pełni kwitnienia (dla szałwii w pełni wzrostu liści). W ostatniej z wymienionych faz oznaczono koncentrację olejku eterycznego. Na podstawie analizy wariancji wykazano, iż oprysk kontrolny miał istotny wpływ na plon suchej masy. Natomiast zawartość olejków eterycznych była najbardziej uzależniona od dwukrotnego oprysku preparatem „Multivit plus" (różnice statystycznie istotne).
Lemon balm, garden sage and common thyme are herbal species possessing versatile medicinal properties. They are also used as spices for culinary purposes. In the Polish climate, despite being perennial, these herbs usually grow on plantations for a year. The reason is their tendency to develop more lignified lower parts of stems, which in consequence depreciates the quality of yield and make the plants more sensitive to spring frost when there is no snow cover. The study aimed at evaluating the yielding of three herbal species (lemon balm, garden sage and common thyme) grown from seedlings in south-eastern Poland. Another objective was to determine the content of essential oils and minerals in raw material against the backdrop of the weather conditions during the growing season. The plant height was determined before harvest. Herbs were cut at the beginning of flowering, i.e. mid-July (lemon balm) and mid-August (common thyme); garden sage does not flower in the first year, hence the herb was cut at the end of August. Weight of fresh herbage was determined after the harvest, while air-dried herbage weight, ground herbage weight (for common thyme) and air-dried leaf weight (for lemon balm and garden sage) were assessed after drying under natural conditions. The content of essential oils was evaluated in dried material according to the applicable method descibed in Pharmacopoeia. Quantities of total nitrogen, ammonia, nitrates, phosphorus, potassium, calcium, magnesium, and sulfur were also determined in the harvested material. The height of lemon balm, garden sage and common thyme plants depended on the rainfall sum. Herbs grew higher in the years characterized by larger sums of atmospheric precipitations from May till July. Rainfalls also significantly affected the yields of fresh lemon balm, garden sage and common thyme herbs. In 2006, a year with less atmospheric precipitation, considerably more total nitrogen, phosphorus, potassium, calcium, magnesium, and sulfur accumulated in lemon balm leaves. More total nitrogen in garden sage and common thyme, ammonia in garden sage leaves as well as calcium in common thyme herb were found as well.
Shoot tips excised from shoot culture of Salvia officinalis were encapsulated in 2% or 3% (w/v) sodium alginate and exposed to 50 mM calcium chloride for complexation. Immediately or after 6, 12 or 24 weeks of storage at 4°C, the synthetic seeds were cultured for 6 weeks on half-strength MS medium supplemented with indole-3- acetic acid (IAA) (0.1 mg/l) and solidified with 0.7% agar. The frequency of shoot and root emergence from encapsulated shoot tips was affected by the concentrations of sodium alginate and additives in the gel matrix (sucrose, gibberellic acid, MS nutrient medium) as well as duration of storage. The frequency of shoot and root induction of non-stored synthetic seeds was highest with shoot tips encapsulated with 2% sodium alginate containing 1.5% sucrose and 0.5 mg/l gibberellic acid (GA3). Shoot tips maintained their viability and ability to develop shoots even after 24 weeks of storage when they were encapsulated in 3% alginate with 1/3 MS medium, sucrose (1.5%) and GA3 (0.25 mg/l). Root formation tended to decrease with storage time. Overall, 90% of the plantlets derived from stored and non-stored synthetic seeds survived in the greenhouse and grew to phenotypically normal plants. This procedure can enable the use of synthetic seed technology for germplasm conservation of S. officinalis, a plant species of high medical and commercial value.
