Organic extracts of six urine samples from children treated with nitisinone, a medicine against tyrosinemia type I, were investigated by 1H and 19F NMR spectroscopy. The presence of unchanged 2-[2-nitro-4-(trifluoromethyl)benzoyl]cyclohexane-1,3-dione (NTBC), 6-hydroxy-2-[2-nitro-4-(trifluoromethyl)benzoyl]cyclohexane-1,3-dione (NTBC-OH) and 2-nitro-4-trifluoromethylbenzoic acid (NTFA) as well as a few other unidentified compounds containing CF3 group was documented.
A heteropolysaccharide, named L2, from Lentinula edodes has been proved to possess immunostimulating and anti-ageing activities in previous studies, but its acting mechanism was not completely understood. In this study, 1 H NMR spectroscopy approach was employed to investigate the metabolic profi les of the urine from adult mice after L2 intervention. Using principal component analysis (PCA) and partial least squares-discriminate analysis (PLS-DA), 22 potential biomarkers were found to be mainly involved in some metabolic pathways: amino acid metabolism, energy metabolism, lipid metabolism, tricarboxylic acid (TCA) cycle, urea cycle and gut microbiota metabolism. Among them, the signifi cantly altered metabolites include: elevated glutamate (75%) and creatine (64%); decreased proline (65%), betaine (58%), fucose (63%) and dimethylamine (59%). In conclusion, the present data is helpful to understand the mechanisms related to previously confi rmed immunomodulation and anti-aging effects of L2, and provide valuable information for mining new functions of L2.
In the solution structure of the ribosome-associated cold shock response protein Yfia of Escherichia coli in the free state two structural segments can be distinguished: a well structured, rigid N-terminal part displaying a βαβββα topology and a flexible C-terminal tail comprising last 20 amino-acid residues. The backbone dynamics of Yfia protein was studied by 15N nuclear magnetic relaxation at three magnetic fields and analyzed using model-free approach. The over all diffusional tumbling of the N-terminal part is strongly anisotropic with a number of short stretches showing increased mobility either on a subnanosecond time scale, or a micro- to milli second time scale, or both. In contrast, the unstructured polypeptide chain of the C-terminal part, which cannot be regarded as a rigid structure, shows the predominance of fast local motions over slower ones, both becoming faster closer to the C-terminus.
