The study was investigated the effects of 10-day treatment with chelidonine (50 and 100 mg/kg ip) on the concentration of creatinine and urea in the blood serum of rats subacutely intoxicated by copper. Subacute intoxication with copper chloride has been found to cause a significant decrease in the urea level and no effect 011 the creatinine level. Chelidonine had virtually 110 effect on the parameters examined except for the decrease in urea in the dose of 50 mg/kg as compared with the control group. 10-day treatment with both doses of chelidonine significantly decreased the level of creatinine and increased the level of urea (dose 100mg/kg) in the serum of intoxicated rats.
W pracy wykazano, że alkaloidy Chelidonium majus L. t.j. allokryptopina, protopina, chelidonina i sangwinaryna (w dawkach odpowiadających 0,025, 0,05, bądź 0,1 LD50), nie wywierały działania przeciwdrgawkowego w maksymalnym elektrowstrząsie u myszy. Co więcej, alkaloidy te nie wpływały na ochronne działanie leków przeciwdrgawkowych. Wyjątek stanowiła chelidonina, która podana łącznie z luminalem, bądź hydantoiną, nasilała ich przeciwdrgawkowy efekt zmniejszając ich ED50.

The behavioral and biochemical studies showed that daily oral administration ol sanumgerman (340 mg/kg, i.e. 1/5 LD₅₀ ) for 3 days exerted a depressive influence on the central nervous system of the mouse. The mechanism of this action could be related to an inhibition of cate- cholaminergic and of serotoninergic central system activities.
In in vitro study on green monkey kidney (GMK) cell culture, the cytotoxicity of Ukrain, methotrexate (MTX), and their combination was investigated. The effect of these drugs and their combination on viability (MTT test) and apoptosis of the cells was assessed. The IC₁₀ and IC₅₀ values for Ukrain and MTX were also indicated. After 24 h of incubation of GMK cells with Ukrain, IC₁₀ amounted to 84.6 µmol/L and IC₅₀ was 256.3 µmol/L, while MTX IC₁₀ amounted to 7.18 µmol/L and IC₅₀ was 154.8 µmol/l. After 24 h of simultaneous incubation of GMK cells with 50 µmol/L of Ukrain and 5.5 µmol/L of MTX, 15.33 % of cytotoxicity of the drugs in LDH test was found. The most significant increase in the cytotoxicity (42.10 %) was noted after 24 h incubation of GMK cells with 150 µmol/L of Ukrain and 5.5 µmol/L of MTX. Likewise, in the MTT assay the greatest decrease in the cells viability was found after incubation with 150 µmol/L of Ukrain and 5.5 µmol/L of MTX. The evaluation of apoptosis also indicated the adverse effect of combined application of both drugs on GMK cells.
Prezentowana praca jest kontynuacją badań nad wpływem wybranych inhibitorów ADH i MEOS na pierwszy etap biotransformacji metanolu. Stosowano: 3-amino-1,2,4-triazol, cymetydynę, EDTA, 1,10-fenantrolinę i 4-metylopirazol w reakcji utleniania metanolu w stężeniach 0,05 i O, 10 mol/L Oceniano ich wpływ na aktywność katalazy w homogenatach wątroby szczurów.
In this study we have investigated the effects of 10-day treatment with chelidonine or Ukrain on the activities of aminotransferases (ALT and AST) in the serum of copper-poisoned rats. It was found that the acute copper chloride poisoning significantly enhanced AST and ALT activity in serum of rats in comparison in the control group. 10-day treatment with chelidonine (50.0 and 100.0 mg/kg i.p.) or Ukrain (7.0 and 14.0 mg/kg i.p.) significantly increased the activity of AST and ALT (only for Ukrain in a dose of 7.0 mg/kg) in the serum of cooper-poisoned rats in comparison with the group receiving cooper chloride only. The results indicate that 10-day treatment with chelidonine or Ukrain increased the activities of aminotransferases in serum of copper-poisoned rats and did not affect the protective changes.
Badano wpływ ostrego i 10-dniowego podawania ip alkaloidów protopinowych: allokryptopiny protopiny na aktywność aminotransferaz ALT i AST, a także oznaczano poziom a-fetoproteiny (AFP) oraz ß2-mikroglobuliny w surowicy krwi szczurów. Stwierdzono, że protopina zastosowana szczurom jednorazowo zwiększała, bądź zmniejszała aktywność AST. Protopina podawana przez 10 dni istotnie zwiększała, aktywność AST i pozostawała bez wpływu na aktywność ALT. Allokryptopina podana jednorazowo w dawce 0,1 LD50 istotnie zmniejszała, natomiast podawana przez 10 dni pozostawała bez wpływu na aktywność obu aminotransferaz. Oba badane alkaloidy nie wpływały na poziom AFP i ß2-mikroglobuliny.
W pracy badano efekty ostrego i 10-dniowego podawania ip alkaloidów benzofenantrydynowych, tj. chelidoniny i sangwinaryny na aktywność aminotransferaz (ALT i AST) oraz oznaczano poziom a-fetoproteiny (AFP) i ß2-mikroglobuliny w surowicy krwi szczurów. Wykazano, że ostre, bądź 10-dniowe podanie chelidoniny nie wpływało na aktywność obu badanych aminotransferaz. Sangwinaryna podana w dawce 0,1 LD50 jednorazowo nie zmieniała aktywności ALT, ale istotnie zmniejszała aktywność AST. Natomiast zastosowana przez 10 dni istotnie zwiększała aktywność AST we wszystkich stosowanych dawkach. Uzyskane podwyższenie aktywności aminotransferaz w surowicy krwi szczurów poddanych działaniu sangwinaryny może sugerować hepatotoksyczne działanie tego alkaloidu. Oba badane alkaloidy zarówno po jednorazowym, jak też 10-dniowym podaniu, nie wpływały na poziom AFP i ß2-mikroglobuliny w surowicy krwi szczurów.
The aim of the study was to investigate the effect of 10-day treatment with Padma 28 on the levels of α-fetoprotein (AFP) and β₂-microglobulin in the serum of cadmium-poisoned rats. It was found that subacute intoxication with cadmium chloride significantly enhanced AFP and β₂-micro-globulin levels in the serum of cadmium-poisoned rats in comparison with the group receiving cadmium only. The data presented here indicated protective action of Padma 28 in cadmium-poisoned rats.
The aim of the study was to investigate the effect of 10-day treatment (i.p.) with Padma 28 on the levels of a-fetoprotein and ß2-microglobulin and the activities of aminotransferases ALT and AST in the serum of cadmium-poisoned rats (dose of 0.36 mg/kg i.p.). It was found that acute intoxication with cadmium chloride significantly enhanced AFP and ß2-microglobulin levels and did not affect the activity of ALT and AST in the serum of rats in comparison with the control group. Padma 28 administrated for 10-days only in the dose of 1 mg/kg significantly decreased AFP and in the doses of 0.1 and 1 mg/kg increased ß2-microglobulin levels while it did not affect the activity of ALT and AST in the serum of cadmium-poisoned rats. 10-day treatment with Padma 28 has been found to increase nephrotoxic effects of cadmium in rats acutely intoxicated with this metal.
