The activities of six enzymes associated with carbohydrate metabo­lism were measured both in carcinomas and in normal breast tissues. The following differences were observed. 1. The carcinoma showed higher enzyme activities than the normal nummary tissue. 2. The ratios of glutamate dehydrogenase, hydroxybutyrate dehydrogenase, gluta­thione reductase and catalase to lactate dehydrogenase were lower in carcinomas than in normal tissues. Similarly, the ratios of glutamate dehydrogenase, hydroxybutyrate dehydrogenase, glutathione reduc­tase and catalase to glucose-6-phosphate dehydrogenase were also sig­nificantly lower in carcinomas. 3. There were no significant differences in enzyme activities between I and II stage of the disease and the metastatic tissues, however, there were significant differences between I and III stage. The significance of these findings is discussed in terms of the alterations in the balance between the metabolic pathways.
This study aimed to determine the changes in the expression of genes for selected specific transcriptional factors that have both activating and repressing functions in in vitro ductal breast cancer cells, under the influence of paclitaxel, applying the microarray technique. The cells are treated with 60 ng/ml and 300 ng/ml doses of paclitaxel that correspond to those applied in breast cancer therapy. About 60 ng/ml doses of paclitaxel cause a statistically significant increase in expression of all the 16 analysed genes coding transcriptional factors, ranging from 1.84-fold (for PO4F2) to 4.65-fold (for LMO4) (p < 0.05) in comparison with the control cells, and enhanced the taxane mechanism of action. The 300 ng/ml doses of paclitaxel cause a cytotoxic effect in the cells. In this article, we argue that these changes in gene expression values may constitute prognostic and predictive factors in ductal breast cancer therapy.
Superoxide dismutase (SOD) plays an important role in detoxication of the organism. Its function is to protect the organism against the cytotoxic action of free radicals. The highest expression of superoxide dismutase was observed in the hepatocytes adjacent to the hydatid cyst. The expression of this gene in the hepatocytes of infected livers 5 cm apart from the infection site was slightly lower.
W pracy zajmowano się grzybem nadrzewnym - modrzewnikiem lekarskim (Laricifomes officinalis). Z owocnika i mycelium officinalis oraz z grzybni hodowanego Lentinus edodes przygotowano frakcje i badano ich aktywność biologiczną w hodowlach komórek nowotworowych HeLa S3. Frakcje te przejawiały zróżnicowane działanie na badane hodowle. Stwierdzono, że najbardziej zaznaczone działanie cytotoksyczne wykazuje frakcja eterowa, natomiast kwas agarycynowy pobudza rozwój komórek linii HeLa.
The investigations concerning the spirochaetes Borrelia burgdorferi sensu lato were conducted in 169 Ixodes ricinus female ticks (141 collected from roe deer and 28 from red deer) as well as in 11 Dermacentor reticulatus female ticks from deer shot in the South-East of Poland. Moreover, a single female Dermacentor reticulatus from a dog and 9 collected from plants were examined. DNA was isolated using the Genomic DNA Prep Plus kit. Its amplification was conducted by means of the diagnostic PCR-Borrelia kit. The presence of spirochaetes was observed in 31.3% of ticks collected from male roe deer, 21.4% - from female roe deer, and 32.1% - from red deer. The highest numbers of infected ticks were detected to date in the Lublin region. Borrelia burgdorferi was found in 5 (45.4%) Dermacentor reticulatus from deer and in single ticks from a dog and plants. These are the first cases in Poland, which indicate that Dermacentor reticulatus is involved in the transmission of borreliosis.
Objectives: Molecular mechanism of carcinogenesis associated with high risk (HR) type HPV is related to the activity of virus oncoproteins E5, E6 and E7. Their task is to bring the cells to a state enabling synthesis of viral DNA, copying viral particles and promotion of uncontrolled cell growth. Proliferative factors of a feminine genital tract epithelium are also sex hormones — estradiol and progesterone. Steroids influence the transcription of genes involved in cell cycle, acting through specific nuclear receptors (ER and PR). The aim of this study was evaluation of the effect of selected concentrations of 17ß-estradiol and progesterone on survival and proliferation of HeLa line cells. Material and methods: The study was done on a transformed HeLa cell line containing integrated genome HPV of type 18. The lines were incubated in the presence of 17ß-estradiol at the concentrations of lx10-4M, lx10-5M, lx10-7M, lx10-8M. Cell survival was determined with the use of 0.5% of water solution of toluidine blue and the cell proliferation rate were evaluated with the use of BrdU Labelling and Detection Kit and the method ELISA (Roche). Results: The obtained results point to 10% toxic influence on HeLa line cells of 17ß-estradiol at high concentrations after 72 h of incubation. The influence of the hormone on proliferation rate was diversified depending on the hormone concentration and time.
