The aim of this study was a comparison of results of the bile acids test and antipyrine test on calves. The experiment was carried out on 10 healthy calves of Black and White breed. In each subject the antipyrine test was performed on day 10 and 20 after birth. The concentration of bile acids in plasma before feeding (fasting - f SKŻ) and 2 hours after feeding (p SKŻ) were also determined. A correlation analysis was used to compare results of the bile acids test with the antipyrine test. The obtained results indicated that the hepatic biotransformation rate of antipyrine in calves increased significantly with age. The concentrations of fasting bile acids in the plasma of 10 day-old calves and 20 day-old calves were not significantly different. Similar results were obtained in the case of postfeeding bile acids concentrations. In contrast, the concentration of bile acids in plasma 2 hours after feeding was significantly (P<0.01) higher (both in 10 day-old and 20 day-old calves) than the concentration of fasting bile acids. It was determined that fasting and postfeeding concentrations of bile acids in plasma and values of metabolic clearance of antipyrine (C1A) were significantly (P<0.01) correlated.
The aim of this study was to determine the „day-night” changes of the antipyrine biotransformation rate in calves. The experiment was carried out on 10 bulls-calves of the black and white breed. During the experiment the animals were kept in uniform environmental condidions and constant light/dark cycle - L/D = 12:12. The antipyrine test was performed on days 10, 20 and 40 after birth. Antipyrine was administered intravenously at a dose of 1000 mg in a 10% sterile solution. Pharmacokinetic parameters of antipyrine were calculated from the elimination curve in two time-intervals (9.00-17.00 - „day”; 21.00-5.00 - „night”), by means of the TopFit 2.0 test. Mean values of half-life and metabolic clearance of antipyrine for all examined calves (n=10) did not differ significantly. Based on results obtained in each calf, the examined population was divided into 2 groups: in 7 calves a decrease of antipyrine half-life and an increase of metabolic clearance during the day were confirmed. The observed changes increased with the age of the examined calves. A different phenomenon was observed in 3 calves - antipyrine elimination from plasma was faster during the night. Also in this case observed changes increased with the age of the calves. The initial concentration of antipyrine and the distribution coefficient did not differ significantly between day and night.
U 13 cieląt (7 jałówek i 6 buhajków) rasy limousine analizowano w pierwszych dziesięciu dniach życia postnatalnego zmiany stężenia w osoczu krwi triglicerydów, cholesterolu całkowitego oraz cholesterolu HDL i LDL. Cielęta przez cały okres badań utrzymywane były przy matkach. Wykazano, że koncentracja triglicerydów w osoczu krwi cieląt korzystających bez ograniczeń ze swobodnego dostępu do siary, a potem mleka matek jest w pierwszych dniach życia postnatalnego nieco wyższa niż u bydła dorosłego i nie wykazuje zróżnicowania w zależności od płci noworodka. Największy wzrost stężenia triglicerydów w osoczu krwi obserwowano w pierwszych trzech dniach życia. Wahania osobnicze w koncentracji triglicerydów u cieląt obu płci były dość znaczne i wynikały najprawdopodobniej ze zróżnicowanych u poszczególnych osobników pór pobierania pokarmu. Wraz z wiekiem cieląt systematycznie wzrastała koncentracja zarówno cholesterolu całkowitego, jak i obu jego frakcji HDL i LDL. Wzrost stężenia cholesterolu HDL w porównaniu z LDL był jednak proporcjonalnie większy. Wraz z wiekiem cieląt stopniowo zmienił się procentowy udział lipoprotein HDL i LDL w transporcie całości cholesterolu znajdującego się w osoczu krwi. Już od 4.–5. dnia życia postnatalnego ponad 50% cholesterolu całkowitego transportowane było przez lipoproteiny HDL.
The aim of this study was to determine the effect of breed on the changes of antipyrine pharmacokinetics in calves receiving high-carbohydrate food. The experiment was carried out on 40 calves (bulls) cross-breed (20 calves - 25% HF and 20 calves 75% HF) 20 animals in the control group and 20 animals in the experimental group. During the experiment the calves from the control group were fed a food ration containing: 250 g of protein, 400 g of carbohydrates and 250 g of lipids. The calves from the experimental group received the high-carbohydrate food: 250 g of protein, 500 g of carbohydrates and 250 g of lipids. The antipyrine test in the experimental group was carried out at 0 and 7 days after administering the food ration with the carbohydrate supplement. The pharmacokinetics of the model drug was calculated based on the plasma concentration (C) versus time (t) curves (TopFit 2.0 software). Statistical analysis was performed by a paired Student-t-test. The increase of carbohydrate content in food rations did not significantly influence the volume of distribution (Vd) of antipyrine in comparison to the control group. We observed, however, a significant (P<0.01) increase of biological half-life, t₀,₅, and a decrease of metabolic clearance (Clm) of antipyrine in calves receiving high-carbohydrate food. The obtained results indicated that calves receiving the high-carbohydrate food were slower metabolizers of antipyrine, but the genotype had no influence on this process (inhibition of activity enzymes of MFO-CYP450 complex) which is evidenced by statistically insignificant differences of pharmacokinetics parameters of antipyrine between 25% and 75% HF cross-breed calves.
