Effects of the phosphorus content in a nutrient solution (15 or 50 mg P dm-3), growing substrate (rockwool or coconut fiber) and the plant growth stage (for roots: 71 or 113 days after transplanting DAT; for leaves: 71 or 92 DAT) on the chemical composition of roots, the phosphorus content in leaves and the expression of genes encoding proteins involved in the transport of phosphorus from the medium to the plant were investigated in tomato cv. Admiro F1 grown in a foil tunnel. A fertigation system without recirculation was used. Regardless of the plant age and growing substrate, tomatoes fertilized with a nutrient solution containing 50 mg P dm-3 had more phosphorus, iron, boron and copper in roots and more phosphorus in leaves. Irrespective of the stage of plant growth and phosphorus level in the medium, the content of almost all macro- and microelements was higher in roots of plants grown in rockwool than in coconut fiber. The stage of plant growth significantly affected the mineral composition of roots as well as the P content in tomato leaves. More phosphorus was stored in roots of younger plants, whereas the phosphorus content was lower in younger than in older leaves. Our analysis of the gene expression showed that transporters encoded by LePT1-LePT4 were involved in phosphate nutrition. Expression of the genes was generally (except LePT4) higher in plants treated by the solution containing 15 mg P dm-3 than in plants treated by 50 mg P dm-3. The expression of genes LePT2, LePT3 in roots of older plants (113 DAT) was generally higher than in young plants.

Diversity of sooty blotch fungi in Poland

Sooty blotch is one of the most common disease of apples in organic orchards in many countries. Results of molecular studies performed in USA indicated approximately 30 different fungi species associated with this disease. Fungi species causing sooty blotch in Northern, Central and Eastern Poland were identified on the basis of morphology and nucleotide sequence of the rDNA internal transcribed spacer region (ITS). A total 245 isolates were collected in spring and early summer in the years 2006–2009 from fruits with visible symptoms of the disease. Isolates were grown on PDA medium and identified on the basis of morphological characters. DNA was extracted from representative isolates and used as matrices for PCR amplification with ITS1F and ITS4 primers. Fragments of amplified rDNA ITS were sequenced. It was found that 66.53% of all isolates causing sooty blotch were species from genera Microcyclosporella, followed by Aureobasidium pullulans – 22.86%, Microcyclospora sp. – 6.12%, Phialophora sessilis – 3.67%, Peltaster sp. and P. fructicola – 0.41%.
Cysteina jest jednym z prekursorów glutationu. Powstaje z siarczku i O-acetylo-L-seryny (OAS) przy udziale liazy O-acetylserynowej (OAS-TL). OAS jest syntetyzowane przez serynową transacetylazę (SAT) z acetylo-CoA i seryny. Celem pracy była ocena strategii tolerancji na ołów u gorczyc (odmian: Nakielska, Małopolska i zakodowanej linii firmy Phytotech) poprzez określenie ekspresji genów kodujących OAS-TL i SAT.
Robinia pseudoaccacia plants grown hydroponically were treated Pb(NO3)2 with 15, 45 mg Pb²⁺ ∙ dm⁻³. After 6, 12, 24, 72 hours of the metal treatment the plants were collected and dissected organs. The plants accumulated and transported to ground part 0.88% and 1.35% of total accumulated lead for the lower and higher dose of Pb²⁺ respectively. The level of GSH was differed and depended on organs, dose and time treatment of Pb²⁺. We investigate (different pattern of expression) expression of RpGSH1 and RpPCS genes in roots. The study showed that glutathione and genes encoded enzymes connected with synthesis of him, plays important role in the process of detoxification in plant.
Six-week old sunflower seedlings, cv. Ogrodowy, were treated with 0, 15, 45 and 60 mg Pb dm-3, and then the content of lead and selected physiological and biochemical parameters were measured. Photosynthesis efficiency, water relations (intensity of transpiration, relative water content (RWC)) and gene-encoding metallothionein were measured three times after 24, 48 and 72 hours of exposure to Pb. The content of glutathione and lead was analysed after 72 hours’ exposure to Pb. Most of the lead uptake was accumulated in the roots, then in the stems and leaves, but when re-calculated per plant dry weight, the uptake of the metal did not depend on the lead dose applied. The highest 60 mg Pb dm-3 treatment was accompanied by a significant decrease in dry weight content. Moreover, most of the lead taken up in these plants was transported to the stems and leaves (23.6% of total lead uptake). The lead doses used in this study did not affect the intensity of photosynthesis, but a decrease in transpiration and relative water content was observed. The glutathione level in the plants varied depending on the organ examined and the Pb concentration in the treatment. The expression of the metallothionein gene HaMT1 was observed in the stems only. These results indicate that the sunflower cultivar Ogrodowy is a promising plant for phytoremediation of lead-polluted soils.
Sooty blotch is a disease of apple and pear caused by a complex of fungi that blemish the fruit surface. Results of molecular studies indicated approximately 30 different fungi species associated with this disease. Apples and pears with symptoms of sooty blotch were collected in summer and early autumn 2006–2010 from trees grown in fungicide non-treated orchards and small gardens located in various regions of Poland. Fungi causing sooty blotch were isolated from fruits and the isolates were divided into six groups, according to their morphological characters. Growth of the fungi colonies were tested on different agar media (PDA, CMA, MEA and Czapek). The ITS region of rDNA from 16 isolates from the first group was amplified by PCR technique and one representative sequence of this isolates was used to alignment in Gene Bank. This isolate was identified as Aureobasidium pullulans and isolates from this group were compared with it on the base of morphological features.
