Badano wpływ jasmonianu metylu (JA-Me) i inhibitorów biosyntezy jasmonianów, ibuprofenu i galusanu propylu, na akumulację chlorofilu i antocyjanów w pędach Clematis pitcheri rosnących w warunkach in vitro na pożywce MS. Doświadczenia prowadzono w 20°C i 25°C. Do pożywki dodawano JA-Me w stężeniu 1 µМ, ibuprofen 1 µM i 10 µМ, galusan propylu 100 µM lub ich mieszaniny. Pędy rosnące w temperaturze 20°C zawierały więcej chlorofilu i akumulowały więcej antocyjanów niż w 25°C. Dodanie do pożywki JA-Me nie wpływało na zawartość chlorofilu w pędach zarówno w 20°C jak i 25°C. JA-Me nie miał wpływu na akumulację antocyjanów w pędach rosnących w 20°C i w niewielkim stopniu obniżał poziom antocyjanów w pędach rosnących w 25°C w porównaniu do kontroli. Zastosowanie inhibitorów biosyntezy jasmonianów znacznie zwiększało zawartość chlorofilu, w porównaniu do roślin kontrolnych i rosnących na pożywce z dodatkiem JA-Me. Równoczesne podanie JA-Me i galusanu propylu zwiększało zawartość chlorofilu w pędach rosnących w 20°C w porównaniu do traktowania samym galusanem propylu. Kiedy JA-Me był podany łącznie z ibuprofenem, pędy rosnące w 25°C zawierały mniej chlorofilu niż na pożywce z samym ibuprofenem. Ibuprofen dodany do pożywki w stężeniu 10 µM stymulował akumulację antocyjanów w pędach w obu temperaturach. Kiedy JA-Me był podany łącznie z ibuprofenem lub galusanem propylu pędy akumulowały mniej antocyjanów niż na pożywkach z inhibitorami podanymi osobno.

The aim of the present work was to study the effect of methyl jasmonate (JA-Me) and inhibitors of biosynthesis of jasmonate, ibuprofen and propylgallate on the accumulation of chlorophyll and anthocyanins in Clematis pitcheri shoots cultured in vitro on the MS medium. The experiment was carried out at 20°C and 25°C. JA-Me at the concentration of 1 µM, ibuprofen at 1 µМ and 10 µM, propylgallate at 100 µM or their mixtures were added to the medium. Plantlets grown at the temperature of 20°C contained more chlorophyll and accumulated more anthocyanins than at 25°C. The addition JA-Me to the medium did not affect the chlorophyll content in the shoots neither at 20°C nor 25°C. JA-Me did nor affect the accumulation of anthocyanins in the shoots growing at 20°C and to a small degree decreased the anthocyanin level in the shoots growing at 25°C as compared to the control. The use of jasmonate biosynthesis inhibitors significantly increased chlorophyll content as compared to the control plants and plants growing on the medium with the addition of JA-Me. Simultaneous addition of JA-Me and propylgallate increased the content of chlorophyll in the shoots growing at 20°C as compared to the treatment with propylgallate alone. When JA-Me was applied together with ibuprofen, shoots growing at 25°C contained less chlorophyll than those growing on the medium with ibuprofen alone. Ibuprofen added to the medium at the concentration of 10 µM stimulated the accumulation of anthocyanins in shoots at both temperatures. JA-Me applied together with ibuprofen or propylgallate shoots accumulated less anthocyanins than shoots on mediums with inhibitors which were added alone.
