Wizualizacja gradientu aktywnej auksyny i rozmieszczenia białek PIN, odpowiedzialnych za wypływ tego hormonu z komórki, jest możliwa dzięki stworzeniu i wykorzystaniu w badaniach roślin transgenicznych. Używając mikroskopu konfokalnego, pośrednio wykorzystując fluorescencję białka reporterowego GFP, lokalizowano gradient auksyny i rozmieszczenie białek chimerycznych PIN1-GFP, PIN4-GFP oraz PIN7-GFP w młodym i różnicującym się transgenicznym kalusie A. thaliana. Materiał wyjściowy stanowiły fragmenty hipokotyli 7-dniowych siewek roślin transgenicznych – DR5rev::GFP, PIN1:GFP, PIN4:GFP oraz PIN7:GFP A. thaliana. Obserwowano zarówno ekspresję DR5::GFP, jak i białek transgenicznych PIN1-GFP i PIN7-GFP tylko w młodych i intensywnie dzielących się komorkach kalusa. Nie stwierdzono zaś ekspresji PIN4-GFP. Stwierdzono apolarną lokalizację białek PIN1-GFP i PIN7-GFP w plazmalemmie komórek kalusa, związaną najprawdopodobniej z brakiem ukierunkowanego krótkodystansowego transportu tego hormonu pomiędzy komórkami kalusa. W 2-tygodniowym transgenicznym kalusie A. thaliana, w którym zaobserwowano proces różnicowania się komórek, zidentyfikowano podwyższony poziom ekspresji DR5::GFP w cytoplazmie, a obecność białek PIN1-GFP w plazmalemmie, po bazalnej stronie komórek miękiszowych ksylemu, co świadczy, iż białka PIN1 są zaangażowane w długodystansowy transport polarny auksyny w grudkach kalusa.

The visualization of auxin gradient and localization of PIN proteins, responsible for the auxin efflux, is possible due to using transgenic plants in the research. The localization of auxin gradient and chimeric PIN1-GFP, PIN4-GFP and PIN7-GFP proteins was studied by monitoring the fluorescence of the reporter gene GFP in intensively dividing and differentiating callus cells obtained from hypocotyl of DR5::GFP and PIN1:GFP, PIN4:GFP, PIN7:GFP A. thaliana. Starting material comprised fragments of hypocotyls of 7-day-old transgenic seedlings DR5rev::GFP, PIN1:GFP, PIN4:GFP, PIN7:GFP of A. thaliana. The explants were grown on fhe sterile callus inducing medium. Expression of DR5::GFP and chimeric PIN1-GFP and PIN7-GFP proteins was observed only in intensively dividing callus. The expression of PIN4-GFP was not detected. An apolar localization of PIN1-GFP and PIN7-GFP proteins in membranes of callus cells was observed, which probably resulted from the nondirected short-distance transport of auxin between cells. It was shown that in 2-week-old transgenic A. thaliana callus, where the cell differentiation was observed, a high expression of DR5::GFP occurred in the cytoplasm and PIN1-GFP proteins were present in the plasmamembrane, at basal side of xylem parenchyma cells.
