W pracy próbowano zwiększyć akumulację wolnych kwasów fenolowych w tkance kalusowej w kulturach wytrząsanych Melittis melissophyllum L. po dodaniu L-fenyloalaniny jako prekursora. Kultury prowadzono w pożywce MS uzupełnionej BAP (1 mg∙dm-3), NAA (0,5 mg∙dm-3), GA3 (0,25 mg∙dm3). L-fenyloalanina była dodawana do pożywki w 14. dniu od założenia hodowli. Ekstrakty metanolowe z tkanek kalusowych były oczyszczane za pomocą SPE. Analizy kwasów fenolowych prowadzono metodą HPLC. Stężenia L-fenyloalaniny 50 i 100 mg na litr pożywki nie miały żadnego znaczącego wpływu na akumulację kwasów fenolowych. Wyższa zawartość prekursora (200 i 600 mg∙dm-3) wyraźnie zwiększyła biosyntezę wolnych kwasów fenolowych w kulturach kalusowych Melittis melissophyllum L. Uzyskane wyniki zostały opracowane statystycznie przy zastosowaniu metody liczb rozmytych dla próbek małolicznych.

The aim of the present investigation was an attempt at increasing the accumulation of free phenolic acids in Melittis melissophyllum L. callus tissue in agitated cultures after the addition of L-phenylalanine as the precursor. Cultures were maintained in the MS medium supplemented with BAP (1 mg∙dm3), NAA (0.5 mg∙dm3), GA3 (0.25 mg∙dm3). L-phenylalanine was added to the medium on the 14th day after inoculation. Methanol extracts from callus tissues were purified by means of SPE. The analyses of phenolic acids were conducted by the HPLC method. Concentrations of L-phenylalanine of 50 and 100 mg per one dm3 of medium had no significant influence on the accumulation of phenolic acids. Higher amounts of the precursor (200 and 600 mg∙dm3) significantly increased the biosynthesis of free phenolic acids in the callus cultures of Melittis melissophyllum L. Results obtained were statistically developed by fuzzy number method for small amount samples.
