Opracowano metodę otrzymywania haploidów owsa metodą izolowanych pylników. Materiał roślinny stanowiło 25 genotypów owsa. Rośliny donorowe rosły w warunkach szklarniowych, a kłosy ścinano, gdy większość mikrospor znajdowała się w fazie późnojednojądrowej. Pylniki wykładano na pożywki różniące się składem mikro-, makroelementów, witamin oraz stężeniami regulatorów wzrostu. Po 6-8 tygodniach kultury określano liczbę tworzących się androgenicznych struktur zarodkowych oraz oceniano żywotność pylników. Po ok. 2-4 tygodniach zregenerowane haploidalne rośliny aklimatyzowano do warunków szklarniowych i podwajano liczbę chromosomów. W wyniku przeprowadzonych doświadczeń stwierdzono, iż indukcja i regeneracja w kulturach pylnikowych owsa jest możliwa, jeżeli przed izolacją pylników wiechy chłodzono w 4°C przez okres 1-2 tygodni, a pylniki indukowano w temperaturze 32°C przez okres 5 dni, a następnie w 28°C, w ciemności, na pożywkach C17 i W14 z dodatkiem 5 mg∙dm-3 2,4-D, 0,5 mg∙dm-3 BAP, 50 mg∙dm-3 L-cysteiny, 500 mg∙dm-3 mioinozytolu i 20 mg∙dm-3 etefonu. Zagęszczenie kultury i płynność pożywki nie miały statystycznie istotnego wpływu na indukcję i regenerację kultur. Spośród 25 testowanych genotypów owsa, 16 regenerowało embriogeniczne struktury kalusowe, z 8 otrzymano regeneranty, a tylko z 2 genotypów podwojone haploidy.

The aim of the study was to elaborate the method for oat haploid production using anther cultures. Donor plants of 25 oat genotypes grew in the greenhouse conditions, and panicles were cut when most of microspores were at the late uninucleus stage. Anthers were placed on media which differed in micro- and macroelements, vitamins and growth regulators. After 6-8 weeks of the culture the number of androgenic structures and vitality of culture were evaluated. Than, 2-4 weeks later, the regenerated haploids were acclimated to the greenhouse conditions and chromosomes were doubled. It was shown that the induction and regeneration of oat anther culture was possible if panicles were cooled at 4°C for 1-2 weeks before the anther isolation, and anthers were induced at 32°C for 5 days, and next at 28°C, in the dark, on C17 and W14 media with 5 mg∙dm-3 2,4-D, 0.5 mg∙dm-3 BAP, 50 mg∙dm-3 L-cysteine, 500 mg∙dm-3 mioinositol and 20 mg∙dm-3 ethephone. Culture density and media fluidity had not statistically significant effect on the culture induction and regeneration. Among 25 tested oat genotypes, 16 regenerated callus structures, 8 regenerated plantlets, but doubled haploids were obtained only from 2 genotypes.
