W pracy przedstawiono wyniki oceny właściwości mutagennych pirenu, benzo(a)- antracenu i ich mieszanin w proporcjach 1:1 oraz 3:1 w zakresie dawek od 2 do 40 µg/płytkę w płytkowym teście Amesa. W obecności frakcji S9, właściwości mutagenne wykazał benzo(a)antracen oraz mieszaniny pirenu i benzo(a)antracenu. Oceniono również właściwości mutagenne ekstraktów frakcji mikrosomalnej i cytozolowej wątroby szczurów po uprzednim podaniu dootrzewnowym badanych WW A. Ekstrakty nie wykazały wpływu na wzrost rewersji mutacji w szczepach bakterii Salmonella typhimurium.

Pyrene (P), benz(a)anthracene (BA) and 1:1 or 1:3 mixtures of P and В A were evaluated for their mutagenic activity in reverse mutation assay with histidine mutants of S. typhimurium celis in the presence of the standard metabolic activation system derived from rat liver (S9), and in the absence of S9 when the mixtures of these compounds were metabolised in vivo after they had been administered intraperitoneally to rats. Pyrene was not found to be mutagenic to tester strains (TA97a, TA98, ТА 100 and ТА 102) in the assay performed with Arocior 1254-induced S9. However, В A induced dose- dependent increase in reverse mutation of three bacterial strains, but did not induce mutations in strain ТА 102. The highest mutagenic response of the histidine mutants was 362 to 998 rev./plate (at doses ranging from 4 to 40 µg/plate) for strain ТА 100. The in vitro results of the assay of pyrene and ВA mixtures mutagenic effect suggest that pyrene at lover doses may enhance the mutagenic activity of ВA and at higher doses it may inhibit BA mutagenic activity. There was no detectable increase in the induction of the reversions in strain ТА 100 by extracts of microsomes and cytosols (0.5-2.0 mg protein/plate) obtained after intraperitoneal treatment of rats with P or BA alone or P/BA mixtures (after in vivo metabolic activation in rats). A single administration at a totai dose of 4.95 mmoi/kg b.w. (about 10 mg/kg) did not induce an evident increase of tester strain reversions for liver extract collection interval of 24 or 48 h after the injection. We may guess that the active metabolites had been eliminated from liver before the livers were removed from the rats.
