Tissue culture conditions were selected to provide the optimal environment for rapid plant multiplication. Plantlets develop under aseptic conditions on the media containing saccharose, mineral nutrients and growth regulators at low irradiance and very high relative humidity. These conditions result in formation of plantlets with abnormal leaf morphology, anatomy and functioning in comparison to seedlings grown under natural conditions. Reduced epicuticular waxes, impaired stomatal function, poorly developed palisade layer with a significant amount of mesophyll air space were observed in plantlets. In addition, their photosynthetic activity may be reduced. When transferred to ex vitro conditions plantlets are subjected to desiccation and their growth is often slow. To minimize the effect of transplantation stress, the plantlets should be acclimatized before exposition to natural conditions.
