Inheritance of triticale alpha-amylase isozymes from wheat and rye parents

A combination of immuno absorption and isoelectric focusing (IEF) was adapted for the study on the inheritance of α-amylase isozymes in tri ticale from its wheat and rye parents. It was shown that the isozyme pattern of tri ticale contained components present in wheat and rye. Quantitative differences between the spectrum of triticale and that of its parents, and in the rate of isozyme staining were observed. Immunosorbtion with concurrent IEF separation showed that complete precipitation of triticale, rye and wheat isozymes occurred when antibodies against triticale α-amylase were used. The triticale did not appear to contain any isozymes not present in either parent. Analogous experiments with antibodies against rye α-amylase showed complete precipitation of rye α-amylases but only partial precipitation of triticale of wheat α-amylases. By a modification of this method, rye anti-a-amylase immunoserum depleted by wheat α-amylase was used to analyze the extracts of the three grains and triticale aleurones used as a model system. Depletion of triticale α-amylases with antiserum against rye a-amylase produced complete precipitation of the low pI isozymes and partial precipitation of the high pIisozymes. The results of this study indicate that triticale α-amylases are simply inherited from its wheat and rye parents. However, there appears to be an interaction between the alien genomes that affect the amount (and/or the specific activity) of some of the isozymes in the triticale.
