In March and April 2010, cysts from the genus Globodera were isolated, from soil attached to two lots of table potatoes originating from Cyprus. Cysts isolated from both soil samples (the sample FF/136/10 taken in March 2010 and the sample FF/285/10 taken in April 2010) revealed characters of two Potato Cyst Nematode species, namely Globodera rostochiensis (Wollenweber, 1923) Skarbilovich, 1959 and G. pallida (Stone, 1973) Behrens, 1975. Results of molecular tests (PCR and PCR-RFLP) confirmed occurrence of both above mentioned nematode species in analysed samples. Sequencing of DNA obtained during PCR amplification and its comparative analysis indicated on occurrence in tested samples of specimen with 89% level of identity and less frequent with about 95% level of identity with G. pallida sequenced available in the National Center for Biotechnology Information (NCBI) Gene Bank. It allowed to recognize that the tested DNA belonged to G. pallida species. Interesting is similarity of tested DNA of Cypriot populations of G. pallida to South American rather than European populations of this nematode.
