Celem badań było porównanie właściwości przeciwutleniających preparatów białek wyizolowanych z gryki odmiany Fagopyrum esculentum Moench oraz hydrolizatów tych białek uzyskanych w wyniku działania pepsyny oraz proteinazy N. Właściwości przeciwutleniające preparatów zbadano oznaczając: zdolność do chelatowania żelaza(II), aktywność wobec kationorodników ABTS i wobec nadtlenków kwasu linolowego. Przeprowadzono także podstawową charakterystykę białek i peptydów otrzymanych preparatów i hydrolizatów białkowych oraz oznaczono zawartość polifenoli ogółem. Zawartość rozpuszczalnych związków peptydowych i polifenoli w hydrolizatach była większa niż w preparatach białek niehydrolizowanych, co prawdopodobnie przyczyniło się do ich lepszej zdolności do chelatowania jonów żelaza(II). Hydrolizaty białkowe otrzymane za pomocą proteinazy N oraz preparaty białek charakteryzowały się mniejszą aktywnością wobec kationorodników ABTS niż hydrolizaty pepsynowe. Preparaty białek wykazywały najlepsze właściwości przeciwutleniające wobec nadtlenków w emulsji kwasu linolowego. Preparaty białkowe hydrolizowane proteinazą N działały w tym układzie reakcyjnym zdecydowanie najsłabiej. Uzyskane wyniki wskazują, że hydrolizaty i preparaty białek gryki mogą być stosowane do żywności jako naturalne składniki o właściwościach przeciwutleniających.

The aim of this study was to compare the antioxidant properties of protein preparations isolated from buckwheat and hydrolysates of these proteins achieved due to the reaction of pepsin and proteinase N. The research material consisted of buckwheat grain of Fagopyrum esculentum Moench cv. from the cultivation in Poland, from which the flour was produced in the laboratory, as well as buckwheat flour available on markets. Both types of flour were subjected to obtain the protein preparations. To produce buckwheat protein hydrolysates, two enzymes were used: pepsin and proteinase N. Hydrolysis of the isolate using pepsin was carried out for 4 h, while proteinase N was applied – for 6 h. Basic characteristics of the resulting protein preparations and hydrolysates included determination of: the content of total nitrogen, soluble peptide compounds, surface aromatic hydrophobicity of protein-origin compounds, and content of total polyphenols. The antioxidant properties of preparations were examined by determining: the ability to chelate iron(II) ions, activity against cation radicals ABTS, and against linoleic acid peroxides. Contents of soluble peptide compounds and polyphenols in protein hydrolysates from buckwheat was higher than in the preparations, which probably contributed to their better ability to chelate the iron(II) ions. The protein hydrolysates produced by means of proteinase N demonstrated lower activity against cation radicals ABTS and chelating properties than hydrolysates obtained by the application of pepsin. Buckwheat protein preparations deactivated the cation radicals ABTS to a lesser extent than pepsin hydrolysates. The process of enzymatic protein hydrolysis resulted in a decrease (up to eight-fold) of the surface aromatic hydrophobicity, wherein there was no any clear influence of the type of enzyme used on the degree of these changes. The best antioxidant properties against peroxides in linoleic acid emulsion were showed by protein preparations having the highest surface aromatic hydrophobicity. Protein preparations hydrolyzed using proteinase N had in such reaction system the weakest efficiency.
